低頻脈沖電磁場在體外通過上調(diào)miR-219-5p促進(jìn)少突膠質(zhì)前體細(xì)胞分化

背景脊髓損傷是一種常見的中樞神經(jīng)系統(tǒng)(central nervous system,CNS)損傷,其病理生理機(jī)制非常復(fù)雜。脊髓損傷損傷可分為原發(fā)性損傷與繼發(fā)性損傷,其中軸突脫髓鞘改變是繼發(fā)性損傷的重要改變之一。少突膠質(zhì)前體細(xì)胞(Oligodendrocyte precursor cells,OPCs)是髓鞘再生的少突膠質(zhì)細(xì)胞(Oligodendrocytes,OLs)的祖細(xì)胞,微小核糖核酸(micro ribonucleic acids,mi RNAs)已經(jīng)被證實(shí)在OPCs的分化過程中扮演著重要角色,低頻脈沖電磁場(Low frequency pulsed electromagnetic field,PEMF)作為一種無創(chuàng)、安全的生物物理治療方式已經(jīng)被證實(shí)在神經(jīng)退行性慢性脫髓鞘疾病中具有改善癥狀的作用,但是其能否影響OPCs分化及其機(jī)制尚不清楚。目的構(gòu)建PEMF,探索PEMF在OPCs分化過程中的作用及機(jī)制。方法首先構(gòu)建PEMF,將OPCs隨機(jī)分為PEMF組和NPEMF組,兩組分別暴露3天,7天,14天和21天后利用細(xì)胞免疫熒光和Western Blot檢測OPCs不同分化階段特異性標(biāo)...
