透明質(zhì)酸水凝膠緩釋雪旺細(xì)胞外泌體修復(fù)坐骨神經(jīng)長(zhǎng)節(jié)段缺損的實(shí)驗(yàn)研究

目的:(1)體外分離新生SD大鼠雪旺細(xì)胞并培養(yǎng),超速離心法提取雪旺細(xì)胞分泌的外泌體并進(jìn)行特異性蛋白鑒定。透射電鏡觀察雪旺細(xì)胞外泌體的粒徑和分布。(2)研究雪旺細(xì)胞外泌體對(duì)神經(jīng)元及雪旺細(xì)胞的生物學(xué)作用。觀察和研究透明質(zhì)酸凝膠緩釋外泌體的生物學(xué)特征參數(shù)。(3)探討透明質(zhì)酸水凝膠包裹緩釋雪旺細(xì)胞外泌體填充PCL神經(jīng)導(dǎo)管修復(fù)大鼠坐骨神經(jīng)長(zhǎng)節(jié)段缺損的形態(tài)學(xué)和功能學(xué)效果。方法:(1)采用0.1%Ⅳ型膠原酶消化法提取原代大鼠雪旺細(xì)胞,并采用DMEM/F12完全培養(yǎng)基培養(yǎng)和純化。采用S100和Sox10進(jìn)行純度鑒定。采用CCK-8試劑盒對(duì)雪旺細(xì)胞的增殖狀態(tài)進(jìn)行檢測(cè)。采用超高速離心法提取雪旺細(xì)胞分泌的外泌體,利用BCA試劑盒檢測(cè)外泌體中蛋白的含量。Western blot法對(duì)雪旺細(xì)胞分泌外泌體表面標(biāo)志物CD63、TSG101、CD9、Rab5和Na+/K+ATP酶進(jìn)行鑒定。(2)構(gòu)建1%濃度的透明質(zhì)酸水凝膠,并將雪旺細(xì)胞外泌體進(jìn)行包裹。體外放置于PBS中采用BCA試劑盒檢測(cè)外泌體的緩釋曲線。應(yīng)用雪旺細(xì)胞外泌體干預(yù)雪旺細(xì)胞和神經(jīng)元,CCK-8試劑盒檢測(cè)外泌體對(duì)雪...
