丙泊酚通過(guò)上調(diào)miR-219-5p抑制肝癌生長(zhǎng)和轉(zhuǎn)移的作用與機(jī)制研究

研究背景原發(fā)性癌是常見(jiàn)的惡性腫瘤,手術(shù)切除是目前治療肝癌最常用和最有效的方法,然而即使是根治性手術(shù)切除的患者,5年的復(fù)發(fā)率仍高達(dá)70%。圍術(shù)期是癌細(xì)胞轉(zhuǎn)移的關(guān)鍵時(shí)間窗,丙泊酚作為腫瘤切除手術(shù)常用的靜脈麻醉藥,研究顯示其具有抑制腫瘤生長(zhǎng)和轉(zhuǎn)移的作用。丙泊酚抑制肝癌的具體作用及機(jī)制目前國(guó)內(nèi)外尚無(wú)明確報(bào)道。mniRNAs在腫瘤發(fā)生發(fā)展過(guò)程中發(fā)揮著重要作用,通過(guò)前期實(shí)驗(yàn)發(fā)現(xiàn),丙泊酚可顯著上調(diào)miR-219-5p表達(dá),miR-219-5p在多種腫瘤中均具有抑癌作用。因此我們探究丙泊酚是否通過(guò)上調(diào)miR-219-5p的表達(dá)抑制肝癌的生長(zhǎng)與轉(zhuǎn)移,并就具體作用機(jī)制作進(jìn)一步探討。研究目標(biāo)1.驗(yàn)證丙泊酚通過(guò)上調(diào)miR-219-5p的表達(dá)抑制肝癌細(xì)胞增殖與侵襲轉(zhuǎn)移。2.探討丙泊酚上調(diào)miR-219-5p抑制肝癌侵襲轉(zhuǎn)移與EMT進(jìn)程的作用機(jī)制,初步探討miR-219-5p的作用靶基因RAB18。3.揭示RAB18在人肝癌細(xì)胞系、肝癌組織及其匹配癌旁肝組織中的表達(dá)水平,及其在肝癌患者預(yù)后評(píng)估的價(jià)值;探討肝癌發(fā)生發(fā)展過(guò)程中RAB18的功能及調(diào)控機(jī)制,以尋求肝癌侵襲轉(zhuǎn)移新的理論依據(jù)。研究?jī)?nèi)容與方法1.揭示丙泊酚通過(guò)...
