含稀土鑭電紡納米纖維膜修復(fù)家兔全層皮膚缺損

【摘要】:目的:探究聚己內(nèi)酯-明膠-氯化鑭(PCL-GE-LaCl_3)電紡納米纖維膜,在修復(fù)家兔全層皮膚缺損模型的作用。方法:利用靜電紡絲技術(shù)制備PCL-GE、PCL-GE-LaCl_3(0.4%組)、PCL-GE-LaCl_3(0.8%組)三種含鑭離子Wt%不同的膜。將27只成年家兔(雌雄不限)用于創(chuàng)面造模,于每只背部?jī)蓚?cè)作3對(duì)兩兩對(duì)稱的2.50.×2.5cm大小全層皮膚缺損創(chuàng)面6個(gè),將每只家兔6個(gè)創(chuàng)面隨機(jī)分為3組:凡士林對(duì)照組,PCL-GE膜組,PCL-GE-LaCl_3實(shí)驗(yàn)組。對(duì)照組覆蓋普通凡士林紗布,PCL-GE組覆蓋PCL-GE膜,實(shí)驗(yàn)組分別覆蓋PCL-GE-LaCl_3(0.4%)膜、PCL-GE-LaCl_3(0.8%)膜。術(shù)后3天、7天、15天每個(gè)時(shí)間點(diǎn),凡士林對(duì)照組,PCL-GE膜組,PCL-GE-LaCl_3(0.4%組)膜、PCL-GE-LaCl_3(0.8%組)的實(shí)際創(chuàng)面數(shù)分別為6、6、8、8,術(shù)后各組各時(shí)間點(diǎn)觀察創(chuàng)面,計(jì)算創(chuàng)面閉合率,取局部組織做病理切片、HE染色,統(tǒng)計(jì)分析。結(jié)果:(1)PCL-GE、PCL-GE-LaCl_3(0.4%組)、PCL-GE-LaCl_3(0.8%組)三)種電紡納米纖維膜,三種纖維膜表面雖存在少量顆粒狀物,但纖維均較為連續(xù)、均一,且呈現(xiàn)出兩種纖維交錯(cuò)分布的結(jié)構(gòu),有較多孔隙。(2)PCL-GE膜的分解峰對(duì)應(yīng)的溫度大于475℃,PCL-GE-LaCl_3(0.4%組)分解峰對(duì)應(yīng)溫度為475℃、PCL-GE-LaCl_3(0.8%組)分解峰對(duì)應(yīng)溫度為400℃,可見隨著鑭離子含量的增大,復(fù)合納米纖維膜的分解起始溫度下降,表明鑭離子加入PCL-GE中降低了復(fù)合納米纖維材料的熱穩(wěn)定性,易于分解。(3)術(shù)中觀察到電紡膜均與創(chuàng)面貼連緊密,呈半透明狀。術(shù)后3d,電紡膜仍與創(chuàng)面貼連緊密,表面干燥,凡士林組仍可見少量滲出。術(shù)后7d,含鑭納米纖維膜表面仍較潔凈,膜下肉芽形成。術(shù)后15d創(chuàng)面基完成上皮化,表面無材料殘留跡象。(4)術(shù)后3天,PCL-GE-LaCl_3(0.4%組)創(chuàng)面閉合合率顯著高于凡士林對(duì)照組,PCL-GE-LaCl_3(0.8%組)閉合率明顯高于凡士林對(duì)照組及PCL-GE組。術(shù)后7天和15天,與凡士林對(duì)照組相比,其他組無明顯差異。結(jié)論:1、PCL-GE-LaCl_3納米纖維膜具有三維立體形態(tài),有較高的孔隙率。2、隨著鑭離子的增多,PCL-GE-LaCl_3電紡膜的熱穩(wěn)定性下降,易于分解。3、PCL-GE-LaCl_3納米纖維膜具有良好的親水性,生物組織相容性。4、含鑭電紡膜能減少創(chuàng)面分泌物,可能具有局部抗炎作用。5、在創(chuàng)面閉合進(jìn)程早期,PCL-GE-LaCl_3(0.8%組)及PCL-GE-LaCl_3(0.4%組)電紡膜可能具有促進(jìn)創(chuàng)面收縮的作用,以PCL-GE-LaCl_3(0.4%)組最明顯。
[Abstract]:Objective: to investigate the effect of polycaprolactone gelatin lanthanum chloride (PCL-GE-LaCl_3) electrospun nanofiber membrane on repairing full-thickness skin defect in rabbits. Methods: three kinds of Wt% membranes containing lanthanum ions, PCL-GE,PCL-GE-LaCl_3 (0.4% group) and PCL-GE-LaCl_3 (0.8% group), were prepared by electrospinning. 27 adult rabbits (male and female) were used to establish wound models. Three pairs of pairwise 2.50. 脳 2.5cm full-thickness skin defect wounds were made on both sides of each back. Six wounds in each rabbit were randomly divided into three groups: Vaseline control group, PCL-GE membrane group and PCL-GE-LaCl_3 experimental group. The control group was covered with ordinary Vaseline gauze, the PCL-GE group was covered with PCL-GE membrane, and the experimental group was covered with PCL-GE-LaCl_3 (0.4%) membrane and PCL-GE-LaCl_3 (0.8%) membrane, respectively. At each time point of 3 days, 7 days and 15 days after operation, the actual number of wounds in Vaslin control group, PCL-GE membrane group, PCL-GE-LaCl_3 (0.4% group) membrane and PCL-GE-LaCl_3 (0.8% group) was 6, 6, 8, 8. the wound closure rate was calculated, local tissue was taken for pathological section, HE staining and statistical analysis. Results: (1) PCL-GE,PCL-GE-LaCl_3 (0.4% group), PCL-GE-LaCl_3 (0.8% group) electrospun nanofiber membranes, although there were a small amount of particles on the surface of the three kinds of fiber membranes, the fibers were continuous and uniform, and showed two kinds of fibers interlaced structure, with more pores. (2) the decomposition peak of PCL-GE film corresponding to the temperature is more than 475C, The corresponding temperature of decomposition peak of PCL-GE-LaCl_3 (0.4% group) is 475 鈩,
