亞低溫對脊髓損傷后反應(yīng)性星形膠質(zhì)細(xì)胞增生的影響

【摘要】:目的:觀察在不同溫度條件下脊髓星形膠質(zhì)細(xì)胞劃痕損傷活化后的形態(tài)和活性改變,以探討亞低溫對脊髓損傷后反應(yīng)性星形膠質(zhì)細(xì)胞增生的影響。方法:體外原代培養(yǎng)新生SD大鼠脊髓星形膠質(zhì)細(xì)胞,以劃痕實(shí)驗(yàn)制備反應(yīng)性星形膠質(zhì)細(xì)胞。亞低溫選擇33℃,細(xì)胞培養(yǎng)48 h。實(shí)驗(yàn)分為對照組、劃痕組、亞低溫組和劃痕+亞低溫組。各組在相應(yīng)的時(shí)間點(diǎn)觀察細(xì)胞形態(tài),采用免疫熒光染色方法檢測Nestin陽性率,MTT比色法觀察細(xì)胞活性,PI染色方法觀察細(xì)胞凋亡程度。結(jié)果:與對照組和亞低溫組相比,劃痕組和劃痕+亞低溫組細(xì)胞胞體肥大,周圍突起增多、延展以及胞漿豐富,細(xì)胞生長率明顯升高。與劃痕組相比,劃痕+亞低溫組細(xì)胞變化減慢,周圍突起減少,細(xì)胞長入劃痕處所需時(shí)間增加,細(xì)胞Nestin陽性率、PI陽性率和細(xì)胞生長率明顯降低,各結(jié)果差異顯著(P0.01)。結(jié)論:劃痕損傷后星形膠質(zhì)細(xì)胞活化為反應(yīng)性星形膠質(zhì)細(xì)胞并會增生,亞低溫明顯抑制脊髓反應(yīng)性星形膠質(zhì)細(xì)胞的活化增生,并可以抑制星形膠質(zhì)細(xì)胞的凋亡。
