脊髓半橫斷損傷對大鼠骨折愈合的影響
[Abstract]:Background: studies have shown that the nervous system significantly affects the healing process of fractures, and that different levels and components of nerve have different effects on callus. Aim: to establish a rat model of spinal cord hemisection injury with fracture, to observe the different fracture of nerve integrity, the different amount of callus and the histomorphological changes during healing, and to explore its effect on the formation of callus. Methods: 48 male SD rats were divided into two groups. Simple fracture model and spinal cord hemisection injury with fracture model were made and right hemisection spinal cord was cut off. On the 14th and 28th day after operation, the tibia was taken in batches to measure wet mass, X-ray was used to observe the healing of fracture, hematoxylin-eosin staining was used to observe the morphological changes of callus tissue, q PCR SYBR GREEN method was used to detect calcitonin gene-related peptide (CGRP) in the broken bone of fracture. Expression of bone morphogenetic protein 2 m RNA. Results & conclusion: (1) in the group of spinal cord hemisection with fracture, the wet mass of the contralateral tibia was greater than that of the affected side (P0.05), which was higher than that of the simple fracture group (P0.05). (2) X-ray showed spinal cord hemisection injury with fracture, the contralateral callus was the largest, the affected side was the second, and the simple fracture callus was the smallest, (3) hematoxylin-eosin staining showed that the contralateral cells were disarranged in the group of spinal cord hemisection injury with fracture 28 days after injury. The fibrous callus in the affected side was dominant, and the arrangement of cells was regular. (4) the expression of calcitonin gene-related peptide (CGRP) and bone morphogenetic protein-2 (BMP-2) in the simple fracture group was higher than that in the spinal cord hemisection injury with fracture group (P0.05). (5) the results suggest that complete innervation is an essential factor in fracture healing.
