【摘要】：背景:H-型血管主要分布在骨骼干骺端,能促進骨量增多,加速骨的形成。目的:觀察骨關節(jié)炎發(fā)生發(fā)展過程中軟骨下骨H-型血管的表達情況。方法:8周齡健康清潔級C57小鼠隨機分為實驗組和假手術組。實驗組小鼠行右側膝關節(jié)內(nèi)側半月板切除誘導骨關節(jié)炎(MMx模型);假手術組小鼠則僅切開關節(jié)囊而不行內(nèi)側半月板切除。術后4周取膝關節(jié)標本進行組織學分析,番紅O快綠染色評估軟骨損傷程度。免疫熒光檢測CD31、Emcn、基質金屬蛋白酶13表達情況,蘇木精-伊紅染色觀察軟骨下骨改變情況,Micro CT分析軟骨下骨骨量的變化。結果與結論:(1)與假手術組比較,實驗組OARSI評分、軟骨下骨CD31、Emcn共熒光、H-型血管、基質金屬蛋白酶13熒光表達及軟骨下骨骨量均顯著升高(P0.05);(2)結果說明,骨關節(jié)炎過程中,軟骨下骨的H-型血管增多,促進軟骨下骨增生及重塑。
[Abstract]:Background: h-type blood vessels are mainly distributed in metaphyseal bone, which can promote bone mass and accelerate bone formation. Objective: to observe the expression of H-type vessels in subchondral bone during the development of osteoarthritis. Methods: 8 weeks old healthy C57 mice were randomly divided into experimental group and sham operation group. The right medial meniscal excision induced osteoarthritis (MMx model) was performed in the experimental group while in the sham-operated group the capsule of the joint was cut but not the medial meniscus was removed. Histological analysis was performed 4 weeks after operation, and the degree of cartilage injury was evaluated by fast green staining. The expression of CD31,Emcn, matrix metalloproteinase 13 was detected by immunofluorescence and the changes of subchondral bone were observed by hematoxylin and eosin staining. The changes of bone mass in subchondral bone were analyzed by, Micro CT. Results and conclusion: (1) compared with sham operation group, OARSI score, CD31,Emcn co-fluorescence, H- type blood vessel, matrix metalloproteinase-13 fluorescence expression and bone mass of subchondral bone in the experimental group were significantly higher than those in the sham operation group (P0.05). (2) the results showed that the H-type vessels of subchondral bone increased during osteoarthritis, and promoted the proliferation and remodeling of subchondral bone.
