EPO治療對(duì)大鼠脊髓損傷后脊髓運(yùn)動(dòng)功能恢復(fù)的實(shí)驗(yàn)性研究

【摘要】:目的:探討人重組促紅細(xì)胞生成素(rh-EPO)在大鼠脊髓損傷(SCI)后運(yùn)動(dòng)功能恢復(fù)中的作用。方法:選取30只200~230g成年雄性SD大鼠,隨機(jī)數(shù)字表法分為三組:假手術(shù)(Sham)組、脊髓損傷(SCI)組和EPO組,每組10只。Sham組大鼠僅行椎板切除術(shù),不傷及脊髓。SCI組和EPO組大鼠行椎板切除和脊髓打擊術(shù)。術(shù)后E PO組連續(xù)7天腹腔注射rh-EPO,SCI組和Sham組采用相同劑量的生理鹽水。應(yīng)用BBB評(píng)分評(píng)估大鼠術(shù)后第1、3、7、14天的后肢運(yùn)動(dòng)功能恢復(fù)情況。HE染色觀察術(shù)后第14天脊髓形態(tài)學(xué)變化、免疫熒光測(cè)定新生神經(jīng)元的增殖情況。結(jié)果:SCI術(shù)后第1、3天SCI組和EPO組之間BBB評(píng)分差異無統(tǒng)計(jì)學(xué)意義;術(shù)后第7、14天EPO組BBB評(píng)分明顯高于SCI組(P0.05,P0.01)。SCI組可見大量膠質(zhì)瘢痕和空洞形成;EPO組上述改變較SCI組減輕。相較于Sham組,SCI組和EPO組新增殖神經(jīng)元數(shù)目逐漸增多,各組間比較差異有統(tǒng)計(jì)學(xué)意義(P0.01)。結(jié)論:大劑量rh-EPO的治療可減輕脊髓內(nèi)的空洞面積、刺激脊髓新生神經(jīng)元的增殖和促進(jìn)大鼠脊髓損傷后后肢運(yùn)動(dòng)功能的恢復(fù)。
[Abstract]:Aim: to investigate the role of recombinant human erythropoietin (rh-EPO) in the recovery of motor function after spinal cord injury (SCI) in rats. Methods: 30 adult male SD rats were randomly divided into three groups: sham operation group (Sham), spinal cord injury (SCI) group and EPO group (10 rats in each group). The rats in Sham group were treated with laminectomy only. No spinal cord injury. Laminectomy and spinal cord attack were performed in SCI and EPO groups. The E PO group was injected intraperitoneally with the same dose of normal saline as Sham group for 7 days after operation. BBB score was used to evaluate the recovery of motor function in the hind limbs of rats on the 1st and 7th day after operation. The morphological changes of spinal cord were observed by HE staining on the 14th day after operation. The proliferation of neonate neurons was measured by immunofluorescence. Results: there was no significant difference in BBB score between SCI group and EPO group on the 1st day after SCI, and the BBB score in EPO group was significantly higher than that in SCI group on 714 days after SCI (P0.05 + P0.01). SCI group). The above changes in EPO group were less than those in SCI group. Compared with Sham group, the number of new proliferative neurons in SCI group and EPO group increased gradually, and the difference was statistically significant (P0.01). Conclusion: the treatment of high dose rh-EPO can reduce the area of syringomyelia, stimulate the proliferation of new neurons in spinal cord and promote the recovery of motor function of hind limbs after spinal cord injury in rats.
