睪酮對膝關節(jié)骨關節(jié)炎去勢雄兔的軟骨修復作用

【摘要】:目的探討睪酮對膝關節(jié)骨關節(jié)炎(KOA)睪丸切除(去勢)雄兔的軟骨修復作用。方法將18只雄兔采用改良Hulth法建立右側KOA模型并去勢,8周時隨機處死6只,確認模型建立成功。將剩余12只兔隨機分為觀察組和對照組,每組6只。觀察組每隔2周肌注十一酸睪酮6 mg/kg,對照組同時肌注等量生理鹽水,共8周(4次)。分組處理第8、12、16周,采用電化學發(fā)光免疫分析法檢測血清睪酮;分組處理第16周末處死,取出右股骨髁,肉眼觀察標本大體情況,采用免疫組化法檢測軟骨組織胰島素樣生長因子1(IGF-1)表達,并進行關節(jié)軟骨Mankin's評分。結果兩組第8周血清睪酮水平比較差異無統(tǒng)計學意義(P0.05),觀察組第12、16周血清睪酮水平均明顯高于對照組(P均0.05)。對照組右膝關節(jié)明顯腫大,關節(jié)軟骨面變薄、粗糙,軟骨表面色澤變暗,潰瘍較深,邊緣骨贅形成明顯。觀察組右膝關節(jié)腫大程度輕于對照組,軟骨表面有光澤,可見小潰瘍和不規(guī)則裂隙。觀察組軟骨組織IGF-1陽性表達率高于對照組,Mankin's評分低于對照組(P均0.05)。結論睪酮可促進KOA去勢雄兔的軟骨修復;其機制可能與促進IGF-1表達有關。
[Abstract]:Objective to investigate the effect of testosterone on cartilage repair of (KOA) orchiectomy (ovariectomized) male rabbits with knee osteoarthritis. Methods the right KOA model was established by modified Hulth method in 18 male rabbits and 6 rabbits were randomly killed at 8 weeks. The remaining 12 rabbits were randomly divided into observation group (n = 6) and control group (n = 6). Testosterone undecanoate 6 mg/kg, was injected intramuscularly every 2 weeks in the observation group and the control group was intramuscularly injected with the same amount of normal saline for 8 weeks (4 times). Serum testosterone was detected by electrochemiluminescence immunoassay (ECLIA) at week 812 ~ (12). The right femoral condyle was removed from the right femoral condyle at the end of the 16th week of treatment. The gross situation of the specimens was observed with naked eyes. The expression of insulin-like growth factor 1 (IGF-1) in cartilage was detected by immunohistochemical method, and the articular cartilage Mankin's score was evaluated. Results there was no significant difference in serum testosterone levels between the two groups at the 8th week (P0.05). The serum testosterone levels in the observation group were significantly higher than those in the control group at the 16th week (P 0.05). In the control group, the right knee joint was obviously enlarged, the articular cartilage was thinned, rough, the color of the cartilage was darkened, the ulcers were deeper and the marginal osteophytes were formed obviously. The right knee joint in the observation group was slightly enlarged than that in the control group, the cartilage surface was lustrous, small ulcers and irregular cracks could be seen. The positive expression rate of IGF-1 was higher and the Mankin's score was lower in the observation group than that in the control group (P < 0. 05). Conclusion testosterone can promote cartilage repair in KOA ovariectomized rabbits and its mechanism may be related to the promotion of IGF-1 expression.
