持續(xù)灌注和靜態(tài)保存對(duì)中性粒細(xì)胞凋亡的影響
[Abstract]:In order to study the effects of continuous perfusion of cardiopulmonary bypass (CPB), intermittent pressure perfusion and cryopreservation on the apoptosis of neutrophils in bronchoalveolar lavage fluid (BALF) and the phagocytosis of alveolar macrophages (AMs) in dogs. To explore the best method of lung preservation in vitro, 30 Mongrel dogs were randomly divided into three groups: cardiopulmonary bypass group, pressure perfusion group and hypothermia preservation group with 10 dogs in each group. Under the condition of maintaining mechanical ventilation, the lungs were removed completely, the isolated lungs were preserved in three different ways, and the bronchoalveolar lavage fluid specimens were collected according to the time points. The apoptosis of neutrophils in the bronchoalveolar lavage fluid was detected by flow cytometry, and the phagocytosis of apoptotic neutrophils by alveolar macrophages was detected by immunohistochemistry. The neutrophil apoptosis in bronchoalveolar lavage fluid (BALF) was significantly decreased in each experimental group with the passage of time in vitro. At each time point, neutrophil apoptosis was the most in cardiopulmonary bypass group, followed by pressure perfusion group. The lowest in cryopreservation group. The macrophages phagocytosis of apoptotic neutrophils in each experimental group showed a decreasing trend with the prolongation of the time in vitro. The phagocytosis of apoptotic neutrophils in cardiopulmonary bypass group was better than that in intermittent pressure perfusion group and cryopreservation group. Continuous perfusion of isolated canine lung by extracorporeal circulation can increase neutrophil apoptosis, promote phagocytosis of apoptotic neutrophil, avoid delayed apoptosis, and reduce ischemia-reperfusion injury in isolated lung tissue. It has better lung protective effect.
