【摘要】：目的:通過建立肥胖性2型糖尿病(T 2 D M)胃轉(zhuǎn)流手術(shù)(G B P)大鼠模型,探討G B P手術(shù)對肝臟糖代謝相關(guān)因子表達(dá)的影響。方法:將1 2只6周齡S D大鼠隨機(jī)分為正常對照組(N C D,n=4)、高脂組(H F D,n=4)和高脂手術(shù)組(H F D-G B P,n=4)。H F D組大鼠經(jīng)高脂飼料喂養(yǎng)4周后,通過腹腔注射鏈脲佐菌素構(gòu)建T 2 D M模型,術(shù)前比較兩組體質(zhì)量、空腹血糖(F P G)、葡萄糖耐受、胰島素敏感性的變化。2周后,構(gòu)建H F D-G B P組胃轉(zhuǎn)流手術(shù)模型,連續(xù)觀察2周,比較手術(shù)前后兩組體質(zhì)量、空腹血糖、血清膽汁酸(B A)、總膽固醇(T C H)、甘油三酯(T G)的變化,并分別提取3組大鼠肝組織的總R N A和蛋白,通過R T-P C R、W e s t e r n印跡法檢測法呢醇X受體(F X R)、磷酸烯醇式丙酮酸羧基激酶(P E P C K)、葡萄糖-6-磷酸酶(G 6 P a s e)、葡萄糖轉(zhuǎn)運(yùn)體2(G L U T 2)和成纖維細(xì)胞生長因子-2 1(F G F-2 1)的表達(dá)水平。結(jié)果:與N C D和H F D組大鼠相比,H F D-G B P大鼠血清總膽汁酸水平顯著升高(p0.0 5;p0.0 1);與N C D組相比較,H F D組的血清總膽固醇和甘油三酯水平明顯升高(p0.0 5;p0.0 5);與H F D組相比,H F D-G B P組大鼠體重下降,血糖含量降低(p0.0 5),血清總膽固醇和甘油三酯水平顯著降低(p0.0 5;p0.0 1),術(shù)后肝臟F X R和G L U T 2 m R N A表達(dá)明顯上調(diào)(p0.0 5),P E P C K、G 6 P a s e和F G F-2 1 m R N A水平表達(dá)也增加;與N C D組相比,H F D和H F D-G B P兩組F X R和P E P C K蛋白表達(dá)明顯升高(p0.0 5)。結(jié)論:在肥胖型T 2 D M大鼠模型中,G B P手術(shù)能有效改善糖代謝;G B P術(shù)后的血清膽汁酸水平明顯升高;F X R、P E P C K、G L U T 2和F G F-2 1的表達(dá)變化可能參與G B P術(shù)后的血糖調(diào)節(jié)過程。
[Abstract]:Objective: to investigate the effect of G B P operation on the expression of hepatic glucose metabolism related factors in obese type 2 diabetes mellitus (T 2 D M) gastric bypass) (G B P) rat model. Methods: twelve 6-week-old SD rats were randomly divided into three groups: normal control group (N C), hyperlipidemic group (H F) and high-fat operation group (H F D-G B PnN 4). H F D group) fed with high fat diet for 4 weeks. The T 2D M model was established by intraperitoneal injection of streptozotocin. The changes of body mass, fasting glucose (F P G), tolerance and insulin sensitivity were compared between the two groups before and after 2 weeks. A gastric bypass model was established in group H F D-G B P and observed for 2 weeks. The changes of body mass, fasting blood glucose and serum bile acid (B A), total cholesterol (T C H), triglyceride (T G) were compared between the two groups before and after operation. The total R N A and protein were extracted from the liver tissues of three groups of rats, and the (P E P C K), of (F X R), phosphoenolpyruvate carboxykinase was detected by RT-P C RW e s t e r n blot. The expression of glucose-6-phosphatase (G6 P a s e), glucose transporter 2 (G L U T 2) and fibroblast growth factor-21 (F G F-21 was detected. Results: compared with N C D and H F D groups, the serum total bile acid level of, H F D-G B P rats was significantly increased (p 0.05 P 0.01). Compared with the N C D group, the serum total cholesterol and triglyceride levels in the, H F D group were significantly higher than those in the, H F D group (p0.05, p0.05). Compared with H F D group, H F D-G B P group decreased body weight, blood glucose content (p0.05), serum total cholesterol and triglyceride levels (p0.05). The expression of F X R and G L U T 2 m R N A was up-regulated (p0.05), P E P C KnG 6 P a s e and F G F-21 m R N A). Compared with N C D group, F X R and P E P C K protein expression in, H F D and H F D G B P groups were significantly increased (p 0.05). Conclusion: in obese T 2D M rat model, G B P operation can effectively improve the serum bile acid level after glucose metabolism; G B P. The changes of the expression of F X RN P E P C KG L U T 2 and F G F 21 may be involved in the regulation of blood glucose after G B P.
