胃轉(zhuǎn)流手術(shù)對糖尿病大鼠肝臟糖代謝相關(guān)因子表達(dá)的影響
[Abstract]:Objective: to investigate the effect of G B P operation on the expression of hepatic glucose metabolism related factors in obese type 2 diabetes mellitus (T 2 D M) gastric bypass) (G B P) rat model. Methods: twelve 6-week-old SD rats were randomly divided into three groups: normal control group (N C), hyperlipidemic group (H F) and high-fat operation group (H F D-G B PnN 4). H F D group) fed with high fat diet for 4 weeks. The T 2D M model was established by intraperitoneal injection of streptozotocin. The changes of body mass, fasting glucose (F P G), tolerance and insulin sensitivity were compared between the two groups before and after 2 weeks. A gastric bypass model was established in group H F D-G B P and observed for 2 weeks. The changes of body mass, fasting blood glucose and serum bile acid (B A), total cholesterol (T C H), triglyceride (T G) were compared between the two groups before and after operation. The total R N A and protein were extracted from the liver tissues of three groups of rats, and the (P E P C K), of (F X R), phosphoenolpyruvate carboxykinase was detected by RT-P C RW e s t e r n blot. The expression of glucose-6-phosphatase (G6 P a s e), glucose transporter 2 (G L U T 2) and fibroblast growth factor-21 (F G F-21 was detected. Results: compared with N C D and H F D groups, the serum total bile acid level of, H F D-G B P rats was significantly increased (p 0.05 P 0.01). Compared with the N C D group, the serum total cholesterol and triglyceride levels in the, H F D group were significantly higher than those in the, H F D group (p0.05, p0.05). Compared with H F D group, H F D-G B P group decreased body weight, blood glucose content (p0.05), serum total cholesterol and triglyceride levels (p0.05). The expression of F X R and G L U T 2 m R N A was up-regulated (p0.05), P E P C KnG 6 P a s e and F G F-21 m R N A). Compared with N C D group, F X R and P E P C K protein expression in, H F D and H F D G B P groups were significantly increased (p 0.05). Conclusion: in obese T 2D M rat model, G B P operation can effectively improve the serum bile acid level after glucose metabolism; G B P. The changes of the expression of F X RN P E P C KG L U T 2 and F G F 21 may be involved in the regulation of blood glucose after G B P.
