十全大補湯對乳腺癌患者術(shù)后早期恢復(fù)及切口引流液血管生成因子的影響

【摘要】:目的:觀察十全大補湯對乳腺癌患者術(shù)后早期恢復(fù)的作用,并探討對切口引流液血管生成因子的影響以及引流液對乳腺癌細(xì)胞MDA-MB-231增殖、遷移和侵襲能力的作用。方法:將61例乳腺癌病例隨機分為對照組30例和治療組31例,兩組術(shù)后均采用西醫(yī)常規(guī)治療,治療組于術(shù)后第1天起服用十全大補湯,連續(xù)7 d;觀察兩組患者治療前后中醫(yī)證候療效、切口引流時間及不同時點引流量,CCK-8法檢測引流液對MDA-MB-231細(xì)胞增殖的影響、Transwell法檢測細(xì)胞遷移、侵襲能力的變化,蛋白質(zhì)芯片法篩選兩組引流液中血管生成因子的表達(dá)。結(jié)果:(1)十全大補湯組中醫(yī)證候療效總有效率為89.66%,高于對照組的63.33%(P0.05)。(2)十全大補湯組切口引流時間短于對照組引流時間,分別為(12.48±2.11)d、(14.23±1.94)d(P0.05)。兩組引流量均呈下降趨勢,其中第7天十全大補湯組引流量低于對照組(P0.05)。(3)引流液對MDA-MB-231細(xì)胞有促增殖作用,十全大補湯組引流液對MDA-MB-231乳腺癌細(xì)胞的促增殖作用低于對照組(P0.05)。(4)Transwell結(jié)果發(fā)現(xiàn)引流液能促進MDA-MB-231細(xì)胞遷移和侵襲能力,而十全大補湯組的引流液促遷移和侵襲能力較對照組降低(P0.05)。(5)人血管生成因子抗體芯片篩選發(fā)現(xiàn)43種血管生成因子在引流液中均有表達(dá),部分促血管生成因子上調(diào)≥2倍,其中十全大補湯組IL-8、Leptin的表達(dá)水平低于對照組(P0.05)。結(jié)論:十全大補湯能改善乳腺癌患者術(shù)后早期氣血兩虛癥候,有效縮短術(shù)區(qū)引流時間、減少術(shù)區(qū)引流量;切口引流液能促進MDA-MB-231的增殖、遷移及侵襲,十全大補湯可能通過降低引流液中IL-8、Le Ptin等促血管生成因子的表達(dá)進一步抑制引流液對MDA-MB-231細(xì)胞的促增殖、遷移及侵襲能力。
[Abstract]:Objective: to observe the effect of Shiquandabu decoction on the early postoperative recovery of breast cancer patients, and to explore the effect on the angiogenesis factor of incision drainage fluid and the effect of drainage fluid on the proliferation, migration and invasion of breast cancer cell line MDA-MB-231. Methods: 61 cases of breast cancer were randomly divided into control group (n = 30) and treatment group (n = 31). After 7 days of treatment, the effects of TCM syndrome, incision drainage time and drainage flow at different time points on the proliferation of MDA-MB-231 cells were detected by CCK-8 method and the changes of cell migration and invasion ability were detected by Transwell method. The expression of angiogenic factor in drainage fluid was screened by protein chip method. Results: (1) the total effective rate of TCM syndromes in the Shiquanduanbu decoction group was 89.66, which was higher than that in the control group (63.33%, P0.05). (2). The drainage time of incision in the Shiquanduanbu decoction group was shorter than that in the control group, which was (12.48 鹵2.11) d, (14.23 鹵1.94) d (P 0.05). The drainage flow of the two groups showed a downward trend, and on the 7th day, the drainage volume of the 10 Dabu decoction group was lower than that of the control group (P0.05). (3), and the drainage fluid had the effect of promoting the proliferation of MDA-MB-231 cells. The effect of drainage fluid on proliferation of MDA-MB-231 breast cancer cells in Shiquandabu decoction group was lower than that in control group (P0.05). (4) Transwell. The results showed that the drainage fluid could promote the migration and invasion of MDA-MB-231 cells. Compared with the control group (P0.05). (5), 43 kinds of angiogenic factors were found to be expressed in the drainage fluid, and some angiogenic factors were up-regulated more than 2 times, compared with the control group (P0.05). (5). The expression level of IL-8,Leptin in Shiquandabu decoction group was lower than that in control group (P0.05). Conclusion: Shiquandabu decoction can improve the symptoms of deficiency of qi and blood, shorten the drainage time and reduce the drainage flow in the early postoperative stage of breast cancer patients, and the drainage fluid of incision can promote the proliferation, migration and invasion of MDA-MB-231. Shiquandabu decoction could further inhibit the proliferation, migration and invasion of MDA-MB-231 cells by reducing the expression of angiogenic factors such as IL-8,Le Ptin in drainage fluid.
