竹葉黃酮緩解異氟醚引起的老年大鼠認(rèn)知功能障礙

【摘要】:研究竹葉黃酮對(duì)異氟醚吸入誘發(fā)老年大鼠神經(jīng)細(xì)胞凋亡及認(rèn)知功能障礙的影響.分離培養(yǎng)原代海馬神經(jīng)細(xì)胞,暴露于體積分?jǐn)?shù)為2%異氟醚6 h后,分別給予50、80及100 mg/L竹葉黃酮處理,MTT法檢測(cè)細(xì)胞增殖能力,流式細(xì)胞儀檢測(cè)細(xì)胞凋亡情況.大鼠經(jīng)吸入1.4%體積的異氟醚2 h后,分別于1~5 d連續(xù)腹腔注射25、50、100 mg/kg竹葉黃酮,于第6天進(jìn)行水迷宮實(shí)驗(yàn)檢測(cè)大鼠空間學(xué)習(xí)記憶能力,ELISA法檢測(cè)海馬組織勻漿中的Aβ1-42、TNF-α、INF-γ含量.結(jié)果發(fā)現(xiàn),竹葉黃酮可顯著促進(jìn)海馬神經(jīng)細(xì)胞的增殖,并且抑制異氟醚所致的海馬神經(jīng)細(xì)胞凋亡.與異氟醚單獨(dú)處理組相比較,各濃度竹葉黃酮可顯著降低大鼠逃避潛伏期并增加大鼠過臺(tái)次數(shù),ELISA結(jié)果顯示竹葉黃酮可顯著抑制海馬組織中Aβ1-42、TNF-α及INF-γ的表達(dá).表明竹葉黃酮可增強(qiáng)大鼠抗炎能力,抑制海馬組織中Aβ1-42的產(chǎn)生,從而抑制異氟醚所致的老年大鼠神經(jīng)損傷及認(rèn)知功能障礙.
[Abstract]:To study the effects of bamboo leaf flavonoids on neuronal apoptosis and cognitive impairment induced by isoflurane inhalation in aged rats. Primary hippocampal nerve cells were isolated and cultured. After exposure to 2% isoflurane for 6 h, 50 or 100 mg/L bamboo leaf flavonoids were used to detect cell proliferation, and flow cytometry was used to detect cell apoptosis. Rats were inhaled with 1.4% isoflurane for 2 h, and 25 50 100 mg/kg bamboo leaf flavonoids were injected intraperitoneally at 1 and 5 days respectively. On the 6th day, the spatial learning and memory abilities of rats were measured by water maze test and the content of A 尾 1-42 TNF- 偽 -INF- 緯 in hippocampus homogenate was detected by Elisa. The results showed that bamboo leaf flavonoids could significantly promote the proliferation of hippocampal neurons and inhibit the apoptosis of hippocampal neurons induced by isoflurane. Compared with the isoflurane treatment group, each concentration of bamboo leaf flavonoids significantly decreased the escape latency and increased the number of rats passing through the stage. Elisa results showed that bamboo leaf flavonoids could significantly inhibit the expression of A 尾 1-42 TNF- 偽 and INF- 緯 in hippocampal tissue. The results showed that the flavonoids of bamboo leaves could enhance the anti-inflammatory ability and inhibit the production of A 尾 1-42 in the hippocampus of rats, thus inhibiting the nerve damage and cognitive impairment induced by isoflurane in the aged rats.
