神經(jīng)營養(yǎng)因子3-殼聚糖載體對大鼠運(yùn)動皮層損傷后內(nèi)源性神經(jīng)發(fā)生和運(yùn)動功能的效果

【摘要】:目的觀察神經(jīng)營養(yǎng)因子3(NT3)-殼聚糖載體對大鼠運(yùn)動皮層損傷后的前肢行為學(xué)功能恢復(fù)的效果,并檢測其對損傷區(qū)及側(cè)腦室下區(qū)神經(jīng)干細(xì)胞(NSCs)的增殖和分化的影響。方法 65只Wistar大鼠分為正常對照組(n=7)、單純損傷組(n=29)和NT3-殼聚糖組(n=29)。制作大鼠運(yùn)動皮層吸除損傷性腦損傷模型。NT3-殼聚糖組于手術(shù)后立即植入NT3-殼聚糖載體,單純損傷組不給予任何治療措施。分別于術(shù)后3 d、7 d、14 d、1個月、2個月和3個月,應(yīng)用食物球抓取實驗觀察大鼠前肢功能恢復(fù)情況,應(yīng)用HE染色觀察損傷區(qū)的空腔體積,應(yīng)用免疫熒光染色評價NSCs的增殖和分化。結(jié)果 NT3-殼聚糖組右側(cè)前肢抓取成功率高于單純損傷組右側(cè)(F6.00,P≤0.05)。NT3-殼聚糖組空腔體積均顯著小于單純損傷組(F629.5,P0.001)。在NSCs分化實驗中,NT3-殼聚糖組各時間點損傷區(qū)Brd U細(xì)胞數(shù)均顯著高于單純損傷組(F171.43,P0.001)。在NSCs增殖實驗中,NT3-殼聚糖組Brd U陽性細(xì)胞數(shù)顯著高于正常對照組和單純損傷組(F155.06,P0.001),術(shù)后7 d損傷同側(cè)Dcx陽性細(xì)胞數(shù)顯著高于單純損傷組(F=62.367,P0.001),Brd U/Dcx雙陽性細(xì)胞數(shù)顯著高于正常對照組(F=33.527,P0.001)。結(jié)論 NT3-殼聚糖載體可增加腦損傷所致的側(cè)腦室下區(qū)NSCs的增殖,促進(jìn)損傷區(qū)新生神經(jīng)元的發(fā)生以及大鼠前肢運(yùn)動功能恢復(fù)。
[Abstract]:Objective to observe the effect of neurotrophic factor 3 (NT3)-chitosan carrier on the recovery of behavioral function of forelimb after motor cortex injury in rats, and to detect the effect of neurotrophic factor 3 (NT3)-chitosan carrier on the proliferation and differentiation of neural stem cells (NSCs) in injured and subventricular regions.
