【摘要】：目的探討七氟烷與高濃度氧對老年小鼠海馬半胱氨酸蛋白酶-3(caspase-3)的表達及認知功能的影響。方法健康雄性C57/BL6老年小鼠(14月齡)33只,采用數(shù)字表法將其隨機分為3組:高濃度氧組(HO組,n=11):吸100%(體積分數(shù))氧4h;高濃度氧+七氟烷組(HS組,n=11):吸100%(體積分數(shù))氧+2.5%(體積分數(shù))七氟烷4 h;對照組(Con組,n=11):吸空氣4 h。24 h后采用Morris水迷宮法研究3組小鼠麻醉后連續(xù)5 d找到平臺所用的時間(逃避潛伏期)和第5天去除平臺后,90 s內(nèi)穿過原平臺的次數(shù)。利用蛋白質(zhì)印記(Western blotting)法檢測小鼠海馬caspase-3的表達。結(jié)果與Con組相比,HO組的caspase-3表達差異無統(tǒng)計學(xué)意義(P0.05);HS組的穿越平臺次數(shù)降低(P=0.023),caspase-3表達顯著增加(P=0.036)。結(jié)論吸入100%(體積分數(shù))氧對老年小鼠認知功能無影響,2.5%(體積分數(shù))七氟烷與100%(體積分數(shù))氧同時吸入增加老年小鼠海馬caspase-3的表達,與老年小鼠產(chǎn)生認知功能障礙有關(guān)。
[Abstract]:Objective to investigate the effects of sevoflurane and high concentration oxygen on the expression of cysteine protease-3 (caspase-3) and cognitive function in hippocampus of aged mice. Methods 33 healthy male C57/BL6 mice (14 months old) were used. They were randomly divided into three groups by digital table: high concentration oxygen group (Ho group): 100% oxygen (volume fraction) for 4 hours; high concentration oxygen sevoflurane group (HS group): inhalation of 100% oxygen (volume fraction) for 4 hours; control group (Con group n 11): The Morris water maze method was used to study the time (escape latency) of finding the platform after anesthesia for 5 days and the times of crossing the original platform within 90 seconds after the removal of the platform on the 5th day after inhaling air for 4 h. 24 h. Protein imprinted (Western blotting) method was used to detect the expression of caspase-3 in hippocampus of mice. Results compared with Con group, there was no significant difference in the expression of caspase-3 between the two groups (P0.05). In HS group, the frequency of crossing platform decreased (P0. 023) and the expression of caspase-3 increased significantly (P0. 036). Conclusion inhalation of 100% (volume fraction) oxygen has no effect on cognitive function of aged mice. Both 2.5% (volume fraction) of sevoflurane and 100% (volume fraction) of oxygen simultaneously increase the expression of caspase-3 in hippocampus of aged mice, which is related to cognitive impairment in aged mice.
