TRPM4通道抑制劑9-Phenanthrol在模擬心內(nèi)直視術(shù)下對幼犬心肌的保護(hù)作用

【摘要】:目的:觀察瞬時受體電位M4型(transient receptor potential melastatin 4,TRPM4)通道抑制劑9-Phenanthrol在模擬心內(nèi)直視下對幼犬心肌的保護(hù)作用。方法:選取18只種類相同,年齡小于8周,體重相近且健康的幼犬作為研究對象,隨機(jī)分為9-Phenanthrol組(n=9)和對照組(n=9)。對照組在體外循環(huán)建立前20分鐘給予生理鹽水,9-Phenanthrol組體外循環(huán)建立前20分鐘給予TRPM4通道抑制劑9-Phenanthrol。然后,應(yīng)用ELISA測定并分別對比兩組幼犬在主動脈阻斷后30分鐘(T1)及60分鐘(T2)、主動脈開放后1小時(T3)及2小時(T4)時刻的血清肌酸激酶同工酶MB(creatine kinase isoenzyme MB,CK-MB)以及肌鈣蛋白I(cardiac troponin I,cTn-I)水平的變化,并應(yīng)用免疫組化和HE染色分析TRPM4在各時刻相同部位心肌組織的表達(dá)情況及心肌組織變性程度。結(jié)果:于T1、T2、T3、T4時刻,對照組和9-Phenanthrol組幼犬血清CK-MB及cTn-I呈升高趨勢,而9-Phenanthrol組血清各指標(biāo)明顯低于對照組,差異均具有統(tǒng)計(jì)學(xué)意義(P0.05)。與對照組相比,9-Phenanthrol組幼犬心肌組織破壞較輕;9-Phenanthrol組幼犬心肌組織中TRPM4蛋白的表達(dá)比對照組中表達(dá)有所減弱。結(jié)論:TRPM4通道抑制劑9-Phenanthrol在模擬心內(nèi)直視術(shù)中的應(yīng)用可對缺血再灌注導(dǎo)致的心肌損傷起到一定程度的保護(hù)作用,對未來應(yīng)用于臨床實(shí)驗(yàn)具有一定的價值。
[Abstract]:Aim: to observe the protective effect of 9-Phenanthrol, an inhibitor of transient receptor potential type M _ 4 (transient receptor potential melastatin _ 4 / TRPM _ 4, on myocardium of puppies under simulated intracardiac direct vision. Methods: eighteen puppies of the same species, age less than 8 weeks and similar and healthy puppies were randomly divided into two groups: 9-Phenanthrol group (n = 9) and control group (n = 9). The control group was given TRPM4 channel inhibitor 9-Phenanthroll 20 minutes before CPB. And then. The changes of serum creatine kinase isoenzyme (MB (creatine kinase isoenzyme MBK-MB) and troponin (I (cardiac troponin ItcTn-I) were measured and compared by ELISA at 30 minutes (T1) and 60 minutes (T2) after aortic occlusion, 1 hour (T3) and 2 hours (T4) after aorta opening. Immunohistochemical staining and HE staining were used to analyze the expression of TRPM4 in myocardium at the same time and the degree of myocardial degeneration. Results: at the time of T1T2T3T4, the serum CK-MB and cTn-I of puppies in control group and 9-Phenanthrol group were increased, while the serum indexes in 9-Phenanthrol group were significantly lower than those in control group (P0.05). Compared with the control group, the expression of TRPM4 protein in the myocardial tissue of 9-Phenanthrol group was lower than that in the control group. Conclusion the application of 9-Phenanthrol in simulated open heart surgery can protect myocardial injury induced by ischemia and reperfusion to a certain extent, and has certain value for clinical trials in the future.
