【摘要】：目的評(píng)價(jià)右美托咪定對(duì)脾切除幼鼠外周血T淋巴細(xì)胞增殖及T細(xì)胞亞群的影響。方法健康雄性SD大鼠24只,6周齡,體重130~150g,隨機(jī)分為四組:生理鹽水組(S組)、右美托咪定組(D組)、生理鹽水組+脾切除(SN組)和右美托咪定組+脾切除(SD組),每組6只。S、D組不做外科處理,SN、SD組制備脾切除模型。喂養(yǎng)1周后S組和SN組腹腔注射生理鹽水10ml/kg,D組和SD組腹腔注射右美托咪定50μg/kg。給藥后2h,取血樣,采用MTT法檢測(cè)外周血T淋巴細(xì)胞增殖能力,采用流式細(xì)胞術(shù)檢測(cè)T細(xì)胞亞群即CD4~+、CD8~+細(xì)胞含量,計(jì)算CD4~+/CD8~+值。結(jié)果與S組比較,SN組T淋巴細(xì)胞增殖能力降低,CD4~+、CD8~+含量及CD4~+/CD8~+值降低,D組T淋巴細(xì)胞增殖能力明顯降低(P0.05);與D組比較,SD組T淋巴細(xì)胞增殖能力明顯降低,CD4~+、CD8~+含量及CD4~+/CD8~+值明顯降低(P0.05);與SN組比較,SD組T淋巴細(xì)胞增殖能力明顯降低(P0.05)。結(jié)論右美托咪定對(duì)正常幼鼠和免疫低下幼鼠細(xì)胞免疫均有一定抑制作用,且對(duì)免疫低下幼鼠的抑制作用更強(qiáng)。
[Abstract]:Objective to evaluate the effect of dexmetomidine on T lymphocyte proliferation and T cell subsets in peripheral blood of splenectomy young rats. Methods 24 male Sprague-Dawley rats were aged 6 weeks. The rats were randomly divided into four groups: normal saline group (S group), dexmetomidine group (D group), splenectomy group (SN group) and desmetomidine group (SD group). One week after feeding, normal saline 10 ml / kg D and SD group were intraperitoneally injected with dexmetomidine 50 渭 g / kg / kg in S group and SN group. Two hours after administration, blood samples were collected and the proliferation ability of peripheral blood T lymphocytes was detected by MTT assay. The T cell subsets (CD4 ~ / CD8 ~ cells) were measured by flow cytometry, and the CD4 ~ / CD8~ value was calculated. Results compared with S group, the proliferation ability of T lymphocytes in S group decreased the content of CD4 ~ + CD8 ~ and CD4 ~ / CD8 ~ value of T lymphocyte in D group (P0.05), and the proliferation ability of T lymphocyte in SD group was significantly lower than that in D group (P0.05), and that in SD group was significantly lower than that in D group (P0.05). Compared with SN group, the proliferative ability of T lymphocytes in SD group was significantly lower than that in SN group (P0.05). Conclusion dexmetomidine has a certain inhibitory effect on the cellular immunity of both normal and immunodeficient young rats, and has a stronger inhibitory effect on the immunosuppression of young rats.
