【摘要】：目的:探討兔前交叉韌帶(anterior cruciate ligament,ACL)損傷后殘端血運(yùn)的變化情況,為ACL保殘重建提供進(jìn)一步的依據(jù)。方法:新西蘭大白兔共30只,雌雄不限,隨機(jī)分成5個(gè)組,每個(gè)組6只,其中1組作為空白對(duì)照組,其余各組分別將其右后肢ACL自股骨端切斷,建立ACL損傷動(dòng)物模型。分別于損傷后2周、4周、8周、12周時(shí)處死實(shí)驗(yàn)動(dòng)物,采集ACL殘端標(biāo)本。將標(biāo)本進(jìn)行HE染色組織觀察、免疫組織化學(xué)染色計(jì)算微血管密度。結(jié)果:與空白對(duì)照組相比,損傷后第2周、4周殘端微血管數(shù)量均升高(P0.05),且4周時(shí)高于其他組(P0.05)。損傷后8周微血管數(shù)量開(kāi)始下降,至12周時(shí)降到最低,低于空白對(duì)照組(P0.05)。結(jié)論:1.ACL殘端血運(yùn)隨時(shí)間變化具有明顯差異性,呈現(xiàn)先增高后降低的趨勢(shì)。2.根據(jù)殘端血運(yùn)變化情況提示,越早進(jìn)行保留ACL殘端重建,對(duì)移植物再血管化就越有利。
[Abstract]:Objective: to investigate the changes of stump blood flow after anterior cruciate ligament (ACL) injury in rabbits. Methods: thirty New Zealand white rabbits were randomly divided into 5 groups with 6 rabbits in each group. The first group was used as the blank control group. The other groups cut the right hind limb ACL from the femur to establish the ACL injury animal model. The experimental animals were killed at 2 weeks, 4 weeks, 8 weeks and 12 weeks after injury, and ACL stump specimens were collected. The specimens were observed by HE staining and microvessel density was calculated by immunohistochemical staining. Results: compared with the control group, the number of microvessels in the stump increased at the 2nd week after injury (P0.05), and was higher than that in the other groups at the 4th week (P0.05). The number of microvessels began to decrease at 8 weeks after injury and reached the lowest at 12 weeks, which was lower than that in the blank control group (P0.05). Conclusion: 1. ACL stump blood flow has obvious difference with time, showing a trend of increase first and then decrease. 2. According to the changes of stump blood flow, the earlier the preservation of ACL stump, the more favorable for graft revascularization.
