A型肉毒素復(fù)合低劑量加巴噴丁對(duì)切口痛大鼠脊髓背角神經(jīng)激肽-1受體內(nèi)化的影響

【摘要】:目的評(píng)價(jià)A型肉毒素復(fù)合低劑量加巴噴丁對(duì)切口痛大鼠行為學(xué)及脊髓背角神經(jīng)激肽-1(neurokinin-1,NK-1)受體內(nèi)化的影響。方法雄性SD大鼠,體質(zhì)量280~300 g,6~8周齡。采用數(shù)字表法隨機(jī)分為5組(n=9):對(duì)照組(Control組)、切口痛—鹽水組(Saline組)、切口痛—加巴噴丁組(GBP組)、切口痛—毒素組(Bo NT/A組)、切口痛—毒素復(fù)合加巴噴丁組(G+B組)。Bo NT/A組和G+B組于造模前1 d鞘內(nèi)注射Bo NT/A 0.5U,GBP組和G+B組于造模前30 min鞘內(nèi)注射GBP 50 mg。切口痛模型制備后3 h,每組各隨機(jī)抽取6只進(jìn)行累計(jì)疼痛評(píng)分(cumulative pain scores,CPS)和機(jī)械縮足閾值(paw withdrawal threshold,PWT)的測量;每組各抽取3只,通過免疫熒光技術(shù)測定NK-1受體內(nèi)化情況。結(jié)果與Control組比較,切口后3 h Saline組、GBP組、Bo NT/A組和G+B組右后足CPS升高、PWT降低、脊髓背角NK-1受體內(nèi)化神經(jīng)元數(shù)目上調(diào)(P0.05);與Saline組比較,Bo NT/A組和G+B組CPS降低、PWT升高、脊髓背角NK-1受體內(nèi)化神經(jīng)元數(shù)目下調(diào)(P0.05),GBP組差異無統(tǒng)計(jì)學(xué)意義(P0.05);與Bo NT/A組比較,G+B組CPS降低、PWT升高、脊髓背角NK-1受體內(nèi)化神經(jīng)元數(shù)目下調(diào)(P0.05)。結(jié)論單獨(dú)應(yīng)用低劑量GBP對(duì)大鼠切口痛無效,Bo NT/A復(fù)合低劑量GBP治療切口痛大鼠的術(shù)后疼痛有明顯的協(xié)同作用,療效優(yōu)于單獨(dú)應(yīng)用Bo NT/A,其鎮(zhèn)痛機(jī)制可能與抑制切口痛大鼠脊髓背角NK-1受體內(nèi)化有關(guān)。
[Abstract]:Objective to evaluate the effects of botulinum toxin A (Botulinum A) combined with low dose of gabapentin on the behavior and the internalization of neurokinin-1 (NK-1) receptor of spinal dorsal horn neurokinin-1 in rats with incision pain. Methods male Sprague-Dawley rats with body mass of 280 ~ 300 g / L and 8 weeks of age were used. Five groups were randomly divided into 5 groups by digital table: control group, saline group, incision pain plus gabapentin group GBP group, incision pain toxin group Bo NT / A group, incision pain toxin combined with Bapentin group, incision pain toxin combined with Bappentin group, G group. Bo NT / A group. Group G B and group G B were injected intrathecal injection of Bo NT / A 0.5 U GBP 1 d before model making, and group G B injected 50 mg of GBP 30 min before modeling. At 3 hours after the establishment of incision pain model, 6 rats in each group were randomly selected for measurement of cumulative pain score (pain score) and mechanical foot contraction threshold (paw withdrawal thresholdpain), and 3 rats in each group were selected for the determination of NK-1 receptor internalization by immunofluorescence technique. Results compared with the control group, the CPS of the right hind foot in the Saline group increased and the number of NK-1 receptor internalized neurons in the dorsal horn of spinal cord increased, compared with the Saline group, the CPS of the Bo NTA group and the G B group decreased the PWT increase, compared with the Saline group, the CPS of the Bo NT / A group and G B group decreased the PWT level. There was no significant difference in the number of NK-1 receptor internalized neurons in the dorsal horn of spinal cord (P 0.05), but the number of CPS in group G B was lower than that in group B, and the number of neurons internalized in NK-1 receptor in the dorsal horn of spinal cord was down-regulated (P 0.05). Conclusion low dose GBP alone has a synergistic effect on postoperative pain in rats with incision pain. The analgesic mechanism may be related to the inhibition of NK-1 receptor internalization in the spinal dorsal horn of rats with incision pain.
