【摘要】：目的探討丙酮酸乙酯(ethyl pyruvate,EP)對脾切除術(shù)后老齡大鼠認(rèn)知功能的影響。方法健康雄性SD大鼠84只,18月齡,體重500~600g,采用隨機(jī)數(shù)字表法將其分為四組,每組21只:對照組(C組)、手術(shù)組(S組)、EP溶液組(E組)和溶劑組(R組)。S組、E組和R組行脾切除術(shù),E組腹腔注射EP 40mg/kg,R組腹腔注射等量EP溶液的溶劑,S組和C組腹腔注射等量生理鹽水。于術(shù)前5d、術(shù)后1、3、7d進(jìn)行Morris水迷宮實(shí)驗(yàn),記錄大鼠逃避潛伏期和游泳距離,水迷宮實(shí)驗(yàn)結(jié)束后處死大鼠,采用Western blot和RT-PCR法檢測四組大鼠海馬高遷移率族蛋白1(high mobility group box 1,HMGB1)及晚期糖基化終末產(chǎn)物受體(receptor for advanced glycation end products,RAGE)的蛋白含量和mRNA表達(dá)量。結(jié)果與C組比較,術(shù)后1、3dS組、E組和R組逃避潛伏期和游泳距離明顯延長,海馬HMGB1、RAGE蛋白含量和mRNA表達(dá)量明顯增加(P0.05)。與S組比較,術(shù)后1、3dE組逃避潛伏期和游泳距離明顯縮短,海馬HMGB1、RAGE蛋白含量和mRNA表達(dá)量明顯減少(P0.05)。與術(shù)前比較,術(shù)后1、3dS組、E組和R組逃避潛伏期和游泳距離明顯延長(P0.05)。結(jié)論丙酮酸乙酯可能通過下調(diào)海馬HMGB1及RAGE的表達(dá)改善認(rèn)知功能。
[Abstract]:Objective to investigate the effect of ethyl pyruvate EPV on cognitive function of aged rats after splenectomy. Methods 84 male Sprague-Dawley rats were randomly divided into four groups. 21 rats in each group: control group (group C), operation group (group S), group E (group E) and solvent group (group R, group C). Group S, group E, and group R, respectively, were treated with splenectomy. Group E was injected intraperitoneally with EP of 40 mg / kg 路kg ~ (-1) / L, group S and group C were injected intraperitoneally with the same amount of EP solution, group S and group C were injected intraperitoneally with the same amount of EP solution. Shoot the same amount of saline. Morris water maze test was performed 5 days before operation and 3 days after operation. The escape latency and swimming distance of the rats were recorded. The rats were killed after the water maze test. Western blot and RT-PCR were used to detect the protein content and mRNA expression of high mobility group protein 1(high mobility group box 1 (HMGB1) and advanced glycosylation end product receptor for advanced glycation end products rages in the hippocampus of rats in the four groups. Results compared with group C, the escape latency and swimming distance of group E and group R were significantly prolonged, and the protein content and mRNA expression of HMGB1 + rage in hippocampus increased significantly (P 0.05). Compared with group S, the escape latency and swimming distance in group E were significantly shortened, and the protein content and mRNA expression of HMGB1 rage in hippocampus decreased significantly (P 0.05). Compared with those before operation, the escape latency and swimming distance of group E and group R were significantly prolonged at 1 day after operation (P 0.05). Conclusion Ethyl pyruvate may improve cognitive function by down-regulating the expression of HMGB1 and rage in hippocampus.
