【摘要】：目的通過動物實驗及體外實驗探索飲酒是否促進乳腺癌惡性進展,且這一作用是否與其誘導(dǎo)的慢性應(yīng)激有關(guān)。方法利用小鼠乳腺癌移植瘤模型,體內(nèi)觀察2%乙醇慢性處理的小鼠乳腺癌E0771細胞對體內(nèi)細胞生長轉(zhuǎn)移的影響;利用體外細胞學(xué)實驗觀察0.2%乙醇慢性誘導(dǎo)對小鼠乳腺癌細胞E0771增殖、遷移及非錨定生長能力的影響;同時用分子生物學(xué)方法探索細胞內(nèi)ROS水平及慢性內(nèi)質(zhì)網(wǎng)應(yīng)激蛋白如p-e IF2a、Bip、XBP1-s等蛋白表達水平是否發(fā)生改變。結(jié)果與對照組相比,飲酒組小鼠體內(nèi)腫瘤生長速度加快,轉(zhuǎn)移增加;體外酒精處理可以顯著促進乳腺癌細胞增殖、遷移等惡性生物學(xué)行為,酒精慢性處理可以誘導(dǎo)ROS、內(nèi)質(zhì)網(wǎng)應(yīng)激活化蛋白p-e IF2a、Bip、XBP1-s表達上升。結(jié)論飲酒可能通過誘導(dǎo)乳腺癌細胞內(nèi)質(zhì)網(wǎng)應(yīng)激促進其惡性進展。
[Abstract]:Objective to investigate whether alcohol consumption promotes the malignant progression of breast cancer and whether this effect is related to chronic stress induced by alcohol consumption in animal and in vitro experiments. Methods the effect of 2% ethanol on the growth and metastasis of mouse breast cancer E0771 cells was observed in vivo by using mouse breast cancer transplanted tumor model. The effects of 0.2% ethanol on the proliferation, migration and unanchored growth of mouse breast cancer cell line E0771 were observed by cytological experiments in vitro. At the same time, the cellular Ros level and the expression level of chronic endoplasmic reticulum stress proteins such as p-e IF2a BipBP1-s were investigated by molecular biology. Results compared with the control group, the tumor growth rate and metastasis increased in the drinking group, and the malignant biological behaviors such as proliferation and migration of breast cancer cells were significantly promoted by alcohol treatment in vitro. Chronic alcohol treatment induced an increase in the expression of endoplasmic reticulum stress activating protein p-e IF2aP1-s. Conclusion drinking may promote the malignant progression of breast cancer cells by inducing endoplasmic reticulum stress.
