【摘要】:目的:研究HSP70對骨髓間充質(zhì)干細胞成骨分化的影響及其相關(guān)機制方法:在成骨分化培養(yǎng)基中添加不同濃度熱休克蛋白70(HSP70)對人骨髓間充質(zhì)干細胞(hMSC)進行培養(yǎng)。采用堿性磷酸酶(ALP)活力檢測以及茜素紅染色了解HSP70在成骨分化早期和晚期的影響。采用PCR及WB技術(shù)檢測成骨相關(guān)標志物的表達變化。最后通過基因芯片技術(shù)和通路富集分析研究HSP70促進成骨分化相通路。結(jié)果:實驗結(jié)果顯示HSP70可顯著提高ALP活力,同時促進鈣結(jié)節(jié)的形成,且存在時間依賴性和劑量依賴性。PCR結(jié)果表明HSP70可顯著促進成骨相關(guān)基因RUNX2,OSX和ALP的表達,WB結(jié)果顯示HSP70可顯著促進成骨相關(guān)蛋白SP7,ALP和BMP2的表達。基因芯片和通路富集分析提示ERK信號通路與HSP70促進hMSC的成骨分化關(guān)系密切。結(jié)論:HSP70通過激活ERK信號通路從而促進hMSC的成骨分化。
[Abstract]:Aim: to study the effect of HSP70 on osteogenic differentiation of bone marrow mesenchymal stem cells (BMSCs) and its related mechanisms: human bone marrow mesenchymal stem cells (hMSCs) were cultured with HSP70 in different concentrations of heat shock protein 70 (HSP70). Alkaline phosphatase (ALP) activity and alizarin red staining were used to investigate the effect of HSP70 on early and late osteogenic differentiation. Polymerase chain reaction (PCR) and WB technique were used to detect the expression of osteoblast related markers. Finally, gene chip technology and pathway enrichment analysis were used to study the pathway of HSP70 promoting osteogenic differentiation. Results: HSP70 significantly increased the activity of ALP and promoted the formation of calcium nodules. The results of time-dependent and dose-dependent .PCR showed that HSP70 could significantly promote the expression of osteoblast-associated genes RUNX2OSX and ALP. The results showed that HSP70 could significantly promote the expression of osteoblast-associated proteins SP7, ALP and BMP2. Gene chip and pathway enrichment analysis suggested that ERK signaling pathway was closely related to HSP70 promoting osteogenic differentiation of hMSC. Conclusion: HSP70 promotes osteogenic differentiation of hMSC by activating ERK signaling pathway.
