七氟醚單次和多次暴露對(duì)新生鼠海馬神經(jīng)元結(jié)構(gòu)的影響

【摘要】:目的探討等同時(shí)間不同次數(shù)七氟醚暴露對(duì)新生大鼠海馬CA1細(xì)胞形態(tài)及超微結(jié)構(gòu)的影響。方法出生7d的SD雄性大鼠45只,體重14~18g,隨機(jī)均分為三組:對(duì)照組(C組)、單次七氟醚暴露組(SS組)和多次七氟醚暴露組(TS組)。SS組于出生后第7天吸入1次3%七氟醚6h;TS組于出生后第7、8、9天每天吸入1次3%七氟醚2h,累計(jì)6h;C組在相應(yīng)日齡吸入60%氧氣。三組于出生后第14天灌注取腦,采用HE和尼氏染色觀察大鼠海馬CA1區(qū)錐體神經(jīng)元形態(tài)及數(shù)量變化;同時(shí),透射電鏡下觀察該區(qū)域神經(jīng)元超微結(jié)構(gòu)及突觸后致密物厚度和活性區(qū)長(zhǎng)度。結(jié)果 HE和尼氏染色顯示:與C組比較,SS組和TS組海馬CA1細(xì)胞排列稀疏,神經(jīng)元數(shù)量明顯減少(P0.05);與SS組比較,TS組海馬CA1神經(jīng)元數(shù)量明顯減少(P0.05)。電鏡結(jié)果顯示:與C組比較,SS組和TS組海馬CA1神經(jīng)元亞細(xì)胞器均有不同程度的受損,突觸后致密物厚度明顯變薄,活性區(qū)長(zhǎng)度明顯縮短(P0.05);與SS組比較,TS組神經(jīng)元亞細(xì)胞器損傷更明顯,突觸后致密物厚度更薄,活性區(qū)長(zhǎng)度縮短更嚴(yán)重(P0.05)。結(jié)論七氟醚單次和多次暴露均會(huì)引起新生大鼠海馬CA1區(qū)錐體神經(jīng)元數(shù)量減少及細(xì)胞超微結(jié)構(gòu)的改變,等同時(shí)間多次暴露比單次暴露對(duì)神經(jīng)元形態(tài)的損傷更嚴(yán)重。
[Abstract]:Objective to investigate the effects of different times of sevoflurane exposure on the morphology and ultrastructure of hippocampal CA1 cells in neonatal rats. Methods Forty-five Sprague-Dawley male rats were born at 7 days. Weight 14g / 18g, randomly divided into three groups: control group (C), single sevoflurane exposure group (SS group) and multiple sevoflurane exposure group (TS group). The SS group inhaled 3% sevoflurane once a day on the 7th day after birth. The TS group was inhaled 1 / day on the 7th day after birth. 3% sevoflurane for 2 h and 6 h cumulative for group C inhaled 60% oxygen at the corresponding age. The rats in the three groups were perfused on the 14th day after birth to observe the morphology and number of pyramidal neurons in the hippocampal CA1 region by HE and Nissl staining. The ultrastructure of neurons, the thickness of postsynaptic dense substance and the length of active area were observed under transmission electron microscope. Results HE and Nissl staining showed that compared with group C, CA1 cells in hippocampus of SS group and TS group were sparsely arranged and the number of neurons decreased significantly (P 0.05), and the number of CA1 neurons in hippocampus of TS group was significantly decreased compared with that of SS group (P 0.05). The results of electron microscope showed that the suborganelles of hippocampal CA1 neurons in SS group and TS group were damaged to some extent, and the thickness of postsynaptic dense substance became thinner than that in C group. Compared with SS group, the suborganelle damage of neurons in TS group was more obvious, the thickness of postsynaptic dense substance was thinner, and the length of reactive area was shorter than that of SS group. Conclusion both single and multiple exposures of sevoflurane can decrease the number of pyramidal neurons and change the ultrastructure of hippocampal CA1 in neonatal rats. Multiple exposures of sevoflurane at the same time are more serious than single exposures to the morphology of neurons.
