急性脊髓損傷大鼠移植骨髓間充質(zhì)干細(xì)胞后神經(jīng)功能恢復(fù)及Nogo-A表達(dá)的影響

【摘要】:目的:探討骨髓間充質(zhì)干細(xì)胞BMSCs(Bone Marrow Mesenchymal Stem Cells)經(jīng)大鼠尾靜脈移植對(duì)大鼠急性脊髓損傷SCI(Spinal cord injury)后神經(jīng)功能的恢復(fù)及對(duì)Nogo-A表達(dá)水平的影響。方法:采用全骨髓貼壁方法分離、培養(yǎng)、純化骨髓間充質(zhì)干細(xì)胞,大鼠急性脊髓損傷模型采用改良艾倫法建立。實(shí)驗(yàn)采用成年的雌性SD大鼠96只,隨機(jī)性分成4個(gè)組,24只/組,分別是假手術(shù)組,SCI組,Vehicle組,BMSCs治療組。骨髓間充質(zhì)干細(xì)胞BMSCs移植前,用羧基熒光素二醋酸鹽琥珀酰亞胺酯(carboxyfluorescein Succinimidyl Ester,CFSE)標(biāo)記。BMSCs組于造模成功10分鐘后,自大鼠尾靜脈注射BMSCs懸液1ml(5×106個(gè)BMSCs);vehicle組則注射同等劑量的PBS。分別在術(shù)后第1天、第3天、第7天和第14天采用BBB神經(jīng)行為學(xué)評(píng)分對(duì)神經(jīng)行為功能進(jìn)行評(píng)估,觀察其神經(jīng)行為功能恢復(fù)情況,采用免疫組化及RT-PCR法測(cè)定脊髓Nogo-A表達(dá)水平。結(jié)果:BMSCs大鼠移植BMSCs成功后,可以在脊髓損傷神經(jīng)的周邊組織區(qū)域中,觀察到陽性CFSE染色細(xì)胞;BMSCs大鼠組損傷后第7天和第14天神經(jīng)功能評(píng)估明顯優(yōu)于損傷對(duì)照組、溶劑對(duì)照組;大鼠BMSCs組術(shù)后第3天、第7天和第14天脊髓損傷區(qū)周邊神經(jīng)組織中Nogo-A表達(dá)比較損傷對(duì)照組和溶劑對(duì)照組,均有明顯降低(P0.01)。.結(jié)論:急性脊髓損傷大鼠移植骨髓間充質(zhì)干細(xì)胞,促進(jìn)損傷后大鼠脊髓神經(jīng)為功能的康復(fù),部分機(jī)制與骨髓間充質(zhì)干細(xì)胞抑制或下調(diào)脊髓組織中Nogo-的生成有關(guān)。
[Abstract]:Aim: to investigate the effects of bone marrow mesenchymal stem cell (BMSCs(Bone Marrow Mesenchymal Stem Cells) transplantation via rat tail vein on the recovery of nerve function and the expression of Nogo-A after acute spinal cord injury (SCI(Spinal cord injuryin) in rats. Methods: bone marrow mesenchymal stem cells were isolated, cultured and purified by whole bone marrow adherent method. The rat model of acute spinal cord injury was established by modified Allen's method. 96 adult female Sprague-Dawley rats were randomly divided into 4 groups (24 rats / group), which were treated with BMSCs in sci group, sci group and BMSCs group. Before transplantation of bone marrow mesenchymal stem cells (BMSCs), carboxyfluorescein Succinimidyl Succinimidyl (BMSCs) was labeled with carboxyfluorescein Succinimidyl (BMSCs). The mice were injected with BMSCs suspension 1ml(5 脳 106 BMSCs in caudal vein of mice. The neurobehavioral function was evaluated by BBB neurobehavioral score on the 1st, 3rd, 7th and 14th day after operation, and the recovery of neurobehavioral function was observed. The expression of Nogo-A in spinal cord was measured by immunohistochemistry and RT-PCR method. Results after the successful transplantation of BMSCs, the positive CFSE staining cells were observed in the peripheral tissue of spinal cord injury nerve. The evaluation of nerve function in the positive CFSE staining group was significantly better than that in the control group on the 7th and 14th day after injury, and that in the solvent control group was better than that in the control group. Compared with the control group and the solvent control group, the expression of Nogo-A in peripheral nerve tissue of BMSCs group was significantly decreased on the 3rd, 7th and 14th day after operation. Conclusion: transplantation of bone marrow mesenchymal stem cells in rats with acute spinal cord injury can promote the recovery of spinal cord nerve function after injury, which is partly related to the inhibition or down-regulation of Nogo- production in spinal cord tissue by bone marrow mesenchymal stem cells.
