脂肪來源干細(xì)胞(ADSCs)對光老化HaCaT細(xì)胞的實(shí)驗(yàn)研究

【摘要】:目的:通過體外共同培養(yǎng)ADSCs(Adipose stem cells,ADSCs)與Ha Ca T細(xì)胞,制造光老化模型,觀察Ha Ca T細(xì)胞形態(tài)學(xué)變化,檢測細(xì)胞培養(yǎng)液中抗氧化酶的活性,探討脂肪來源干細(xì)胞(ADSCs)對光老化Ha Ca T細(xì)胞的抗老化作用。方法:設(shè)計(jì)實(shí)驗(yàn)組為ADSCs與Ha Ca T共培養(yǎng)組,培養(yǎng)4 8h后,給予紫外線UVA30 m J/c m照射24小時(shí)后,繼續(xù)培養(yǎng)24h,對照組為單純培養(yǎng)Ha Ca T,紫外線照射同實(shí)驗(yàn)組。觀察Ha Ca T細(xì)胞形態(tài)學(xué)變化,檢測兩組細(xì)胞培養(yǎng)液中乳酸脫氫酶(LDH)、丙二醛(MDA)、超氧化物歧化酶(SOD)活力。結(jié)果:對照組Ha Ca T細(xì)胞細(xì)胞數(shù)較少,細(xì)胞貼壁性較低,細(xì)胞間隙增大,細(xì)胞變圓或不規(guī)則,細(xì)胞體積變小。實(shí)驗(yàn)組較對照組細(xì)胞形態(tài)較好,圓形、不規(guī)則形、懸浮細(xì)胞較少。實(shí)驗(yàn)組SOD、LDH活力明顯高于對照組(P0.05),實(shí)驗(yàn)組MDA含量明顯低于對照組。結(jié)論:研究結(jié)果表明脂肪來源干細(xì)胞(ADSCs)對光老化有治療作用,能夠減輕UVA對Ha Ca T細(xì)胞的損傷,從而為抗光老化提供新的手段。
[Abstract]:Objective: to establish a photoaging model by co-culture of ADSCs(Adipose stem cells and Ha Ca T cells in vitro, observe the morphological changes of Ha Ca T cells, and detect the activities of antioxidant enzymes in the culture medium. To investigate the anti-aging effect of adipose derived stem cells (ADSCs) on photoaged Ha Ca T cells. Methods: the experimental group was designed as ADSCs and Ha Ca T co-culture group. After 48 hours of culture, ultraviolet UVA30 m J / cm was irradiated for 24 hours, and then cultured for 24 hours. The control group was treated with Ha Ca T, UV irradiation alone. The morphological changes of Ha Ca T cells were observed, and the activities of lactate dehydrogenase (LDH), malondialdehyde (MDA) and superoxide dismutase (SOD) in the culture medium of the two groups were detected. Results: in the control group, the number of Ha Ca T cells was less, the adhesion of cells was lower, the intercellular space was enlarged, the cells became round or irregular, and the cell volume became smaller. The shape of cells in the experimental group was better than that in the control group. The activity of MDA in the experimental group was significantly higher than that in the control group (P 0.05), and the content of MDA in the experimental group was significantly lower than that in the control group. Conclusion: the results suggest that ADSCs can be used to treat photoaging and reduce the damage of Ha Ca T cells induced by UVA, thus providing a new way to resist photoaging.
