LINGO-1在脊髓神經(jīng)干細胞分化過程中的表達特征及效應(yīng)

【摘要】:背景:研究LINGO-1在脊髓神經(jīng)干細胞分化過程中的表達及效應(yīng)對調(diào)控神經(jīng)干細胞分化及修復(fù)脊髓損傷有著重要的意義。目的:觀察LINGO-1在脊髓神經(jīng)干細胞分化過程中的表達特征及生物學(xué)作用。方法:體外培養(yǎng)大鼠脊髓神經(jīng)干細胞,并對培養(yǎng)細胞進行Nestin染色干性鑒定及誘導(dǎo)分化多能性鑒定。于分化第0天及第5天進行LINGO-1與Nestin(神經(jīng)干細胞)、β-TubulinⅢ(神經(jīng)元細胞)、GFAP(星形膠質(zhì)細胞)及O4(少突膠質(zhì)細胞)免疫熒光雙染色,觀察LINGO-1的表達特征。體外培養(yǎng)的大鼠脊髓神經(jīng)干細胞分為對照組及干擾組,干擾組脊髓神經(jīng)干細胞感染LINGO-1 sh RNA慢病毒下調(diào)LINGO-1表達,對照組脊髓神經(jīng)干細胞感染Scramble-sh RNA慢病毒,于感染第5天免疫熒光染色檢測神經(jīng)元突起長度。結(jié)果與結(jié)論:(1)脊髓神經(jīng)干細胞可以分化成為神經(jīng)元、星形膠質(zhì)細胞和少突膠質(zhì)細胞;(2)LINGO-1在脊髓神經(jīng)干細胞及其分化的神經(jīng)元、少突膠質(zhì)細胞中表達,但不在星形膠質(zhì)細胞中表達;(3)下調(diào)LINGO-1表達后,干擾組神經(jīng)元突起長度較對照組明顯增長,差異有顯著性意義(P0.05);(4)結(jié)果表明,體外培養(yǎng)的脊髓神經(jīng)干細胞具有多項分化潛能,LINGO-1在脊髓神經(jīng)干細胞及分化早期的神經(jīng)元、少突膠質(zhì)細胞中表達,但不在星形膠質(zhì)細胞中表達。LINGO-1對分化后神經(jīng)元突起生長具有負性調(diào)控作用。
[Abstract]:Background: to study the expression and effect of LINGO-1 in spinal cord neural stem cell differentiation is of great significance in regulating neural stem cell differentiation and repairing spinal cord injury.
