白細(xì)胞介素1β對(duì)破骨細(xì)胞空泡型質(zhì)子泵活性與胞內(nèi)酸化的影響

【摘要】:目的:通過體外實(shí)驗(yàn)研究白細(xì)胞介素1β(IL-1β)對(duì)破骨細(xì)胞空泡型質(zhì)子泵與細(xì)胞內(nèi)pH值的影響,以探討IL-1β促進(jìn)破骨細(xì)胞骨吸收的機(jī)制。方法:骨髓單核巨噬細(xì)胞經(jīng)過誘導(dǎo)成為破骨細(xì)胞后,予以0.1、0.3和0.5μg/L的IL-1β干預(yù)48 h,檢測(cè)破骨細(xì)胞空泡型質(zhì)子泵mRNA和蛋白的表達(dá)以及IL-1β對(duì)破骨細(xì)胞胞內(nèi)酸化的影響,觀察空泡型質(zhì)子泵活性的改變與破骨細(xì)胞骨吸收功能的變化。結(jié)果:研究結(jié)果提示破骨細(xì)胞經(jīng)IL-1β干預(yù)48 h后,空泡型質(zhì)子泵的表達(dá)與活性均顯著增加,并且細(xì)胞內(nèi)pH值下降。破骨細(xì)胞與含有IL-1β的細(xì)胞培養(yǎng)液共同孵育后,其骨吸收能力增強(qiáng),培養(yǎng)液中IL-1β的濃度越高,該作用越趨明顯。結(jié)論:IL-1β可能是通過上調(diào)空泡型質(zhì)子泵的表達(dá)與酶的活性,促進(jìn)氫離子的產(chǎn)生和(或)轉(zhuǎn)運(yùn),引發(fā)破骨細(xì)胞骨吸收活動(dòng)增加,最終導(dǎo)致骨質(zhì)破壞,由此參與骨關(guān)節(jié)炎癥性疾病的發(fā)生。
[Abstract]:Objective: To study the effect of interleukin 1 beta (IL-1 beta) on the vacuolar proton pump and intracellular pH in osteoclast in vitro, in order to explore the mechanism of IL-1 beta to promote bone resorption of osteoclasts. Method: bone marrow mononuclear macrophages were induced to become osteoclasts, and 0.1,0.3 and 0.5 g/L IL-1 beta intervention 48 h were used to detect osteoclast empty cells. The expression of mRNA and protein in the bubble proton pump and the effect of IL-1 beta on the acidification of osteoclasts, observe the changes in the activity of the vacuolar proton pump and the changes of bone resorption function of the osteoclast. Results: the results suggest that after the intervention of 48 h by IL-1 beta, the expression and activity of the vacuolar proton pump are significantly increased, and the pH value in the cell is also observed. When the osteoclast is incubated with the cell culture fluid containing IL-1 beta, the bone absorption capacity of the cell is enhanced, the higher the concentration of IL-1 beta in the culture medium, the more obvious the effect. Conclusion: IL-1 beta may promote the production and (or) transport of hydrogen ions by up regulation of the expression of the vacuolar proton pump and the activity of the enzyme, causing the bone resorption activity of osteoclasts. The increase will eventually lead to bone destruction, which is involved in the occurrence of bone and joint inflammatory diseases.
