【摘要】：目的觀察椎間盤退變患者髓核組織中高溫必需因子A1(HtrA1)的表達(dá)及其與基質(zhì)金屬蛋白酶(MMPs)的關(guān)系。方法 78例接受椎間盤髓核切除患者,其中年輕脊柱骨折患者(改良Thompson分級Ⅰ級)10例,椎間盤退變患者68例(改良Thompson分級Ⅱ級22例、Ⅲ級24例、Ⅳ級22例)。采用real-time PCR和Western blotting法分別檢測椎間盤髓核組織中的Htr A1及MMP-1、3、7、9、13 mRNA和蛋白,并采用Pearson法分析Htr A1和MMP-1、3、7、9、13的相關(guān)性。結(jié)果改良Thompson分級Ⅰ、Ⅱ、Ⅲ、Ⅳ級患者椎間盤髓核組織中HtrA1 mRNA分別為0.56±0.49、0.70±0.45、1.47±0.86、4.10±1.17,蛋白分別為0.16±0.09、0.52±0.07、1.21±0.26、3.10±0.17,兩兩相比P均0.05。不同改良Thompson分級患者椎間盤髓核組織中MMP-1、3、13的mRNA、蛋白表達(dá)兩兩相比P均0.05。HtrA1 mRNA與MMP-1、3、13 mRNA表達(dá)呈正相關(guān)(r~2分別為0.144 8、0.240 6、0.869 5,P均0.01)。結(jié)論椎間盤髓核組織中HtrA1的表達(dá)隨椎間盤退變程度的升高而升高,且與MMP-1、3、13的表達(dá)有關(guān)。
