二甲氨基乙醇和復方氨基酸溶液對衰老大鼠皮膚的Periostin和Dermatopontin表達的影響

【摘要】:目的:根據(jù)中胚層療法(Mesotherapy)理念,觀察皮膚真皮內(nèi)注射二甲氨基乙醇(DMAE)和復方氨基酸溶液(AA)對D-半乳糖(D-gal)誘導的化學性衰老大鼠皮膚的成骨細胞特異性因子(Periostin,POSTN)和皮膚橋蛋白(Dermatopontin,DPT)表達的影響。方法:使用80只Wistar大鼠,入組隨機,按照正常對照組、衰老對照組、衰老治療組。注射D-半乳糖后第18天開始,衰老治療組在大鼠兩側(cè)臀部皮膚分別注射0.1%DMAE+AA、0.2%DMAE+AA、0.1%DMAE、0.2%DMAE、AA、NS,每周1次,連續(xù)4周。觀察各組皮膚組織學變化,真皮厚度、羥脯氨酸(Hydroxyproline,Hyp)、I型及III型膠原蛋白m RNA表達、成骨細胞特異性因子(Periostin,POSTN)和皮膚橋蛋白(Dermatopontin,DPT)表達水平的變化。結(jié)果:在衰老大鼠真皮內(nèi)注射0.1%DMAE+AA、0.2%DMAE+AA,可促進老化大鼠真皮厚度、Hyp含量和I和III型膠原蛋白m RNA及POSTN表達增加,而對DPT表達沒有影響。結(jié)論:在衰老大鼠中注射DMAE和AA復合溶液,能有效改善衰老的皮膚組織結(jié)構(gòu)與已老化且變性的膠原蛋白。同時能是促進衰老大鼠皮膚進行膠原蛋白合成,并明顯增加皮膚真皮厚度及膠原蛋白含量,對抗衰老具有一定延緩老化皮膚的作用。
[Abstract]:Objective: according to the idea of mesodermic therapy, To observe the effects of intradermal injection of dimethylaminoethanol (DMAE) and compound amino acid solution (AAA) on the expression of D-galactose-induced osteoblast specific factor Periostinpost (POSTN) and dermatopontinine (DPTT) in the skin of aged rats. Methods: 80 Wistar rats were randomly divided into two groups: normal control group and aging treatment group. From the 18th day after the injection of D-galactose, the aging treatment group received 0.2DMAE 0.2DMAE 0.2DMAE 0.2DMAEA NSS once a week for 4 weeks in the aging treatment group. The changes of skin histological changes, dermal thickness, the expression of collagen type I and type III collagen m RNA, the osteoblast specific factor Periostin post and the expression level of Dermatopontintinin were observed. Results: injection of 0.1%DMAE AA0.2DMAE into dermis of aging rats increased the content of Hyp and the expression of type I and III collagen m RNA and POSTN in the dermis of aging rats, but had no effect on the expression of DPT. Conclusion: the compound solution of DMAE and AA can effectively improve the aging skin tissue structure and the aging and denatured collagen in aging rats. At the same time, it can promote collagen synthesis in the skin of aging rats, and increase the dermal thickness and collagen content of the skin, which has the effect of delaying aging skin.
