新型大鼠脊髓打擊器的制備及建模大鼠的功能與病理評價

【摘要】:[目的]制備一種簡便、實用的急性大鼠脊髓損傷建模裝置。[方法]用木板、新泡沫板、吸管、手術(shù)縫線及金屬插銷桿等制作新型大鼠脊髓打擊器。取36只成年雌性SD大鼠,體質(zhì)量為220~250 g,隨機(jī)等分為假手術(shù)組(Sham組)、輕度打擊組(SCI1組)和重度打擊組(SCI2組),利用自制大鼠脊髓打擊器建立大鼠脊髓損傷模型,SCI1組采用10 g×25 mm打擊規(guī)格進(jìn)行垂直打擊損傷,SCI2組采用10 g×50 mm打擊規(guī)格進(jìn)行垂直打擊損傷。各組大鼠于術(shù)前1 d,術(shù)后1、3、7、14、21和28 d進(jìn)行BBB運動學(xué)評分,觀察大鼠后肢運動功能情況,術(shù)后28 d對各組大鼠受損脊髓組織進(jìn)行HE染色、神經(jīng)元核抗原(Neu N)免疫組化檢測。[結(jié)果]Sham組各觀察時間點的BBB評分均為21分,各SCI組術(shù)后1 d時BBB評分為0分,之后BBB評分結(jié)果隨時間延長呈逐步恢復(fù)趨勢,從3 d開始,SCI1組的BBB評分高于SCI2組(P0.05)。各組大鼠后肢左右側(cè)活動情況無明顯差異,實驗過程中無大鼠死亡。術(shù)后28 d HE染色、Neu N免疫組化結(jié)果顯示SCI2組脊髓受損程度明顯重于SCI1組(P0.05)。[結(jié)論]該新型大鼠脊髓打擊器制作簡便、可行性高,建立的SCI模型具有穩(wěn)定性高、可重復(fù)性強、損傷程度可調(diào)節(jié)等特點,適合相關(guān)科研人員使用。
[Abstract]:Objective to prepare a simple and practical model device for spinal cord injury in rats. [methods] A new type of rat spinal cord punch was made with plank, new foam plate, suction tube, suture and metal pin. 36 adult female SD rats were selected. The body mass was 220 ~ 250g. The rats were randomly divided into sham-operated group (Sham group), light attack group (SCI1 group) and severe attack group (SCI2 group). The spinal cord injury model of rats was established by using self-made spinal cord batter. The SCI1 group was used 10 g 脳 25 mm strike specification. Direct strike injury group (SCI 2) used 10 g 脳 50 mm strike specification for vertical strike injury. The motor function of hind limbs was observed at 1 day before operation, 1 day before operation and 1 day after operation. The motor function of hind limbs was observed. The injured spinal cord tissues in each group were stained with HE and the neuronal nuclear antigen (Neu N) was detected by immunohistochemistry on the 28th day after operation. [results] the BBB score of Sham group was 21 at each observation time point, and the BBB score was 0 at 1 day after operation in each SCI group, and then the BBB score showed a trend of gradual recovery with the prolongation of time. The BBB score of SCI1 group was higher than that of SCI2 group from 3 days after operation (P 0.05). There was no significant difference in the left and right side movements of the hind limbs in each group, and no rats died during the experiment. On the 28th day after operation, the results of HE staining and Neu N immunohistochemical staining showed that the degree of spinal cord injury in SCI2 group was significantly worse than that in SCI1 group (P 0.05). [conclusion] the new rat spinal cord percussion device is simple and feasible. The SCI model has the characteristics of high stability, strong repeatability and adjustable injury degree, and is suitable for use by related researchers.
