5-氨基酮戊酸光動(dòng)力療法對(duì)瘢痕成纖維細(xì)胞的作用研究

【摘要】:目的研究5-氨基酮戊酸光動(dòng)力療法(ALA-PDT)對(duì)瘢痕成纖維細(xì)胞的作用及其機(jī)制,為臨床運(yùn)用ALA-PDT治療瘢痕疙瘩提供理論依據(jù)。方法取人瘢痕成纖維細(xì)胞原代培養(yǎng),并將生長(zhǎng)狀態(tài)無(wú)明顯差異的瘢痕成纖維細(xì)胞隨機(jī)分為空白對(duì)照組、磷酸鹽緩沖液(PBS)組以及6個(gè)不同濃度的5-氨基酮戊酸(ALA)培養(yǎng)液組。每組給予635 nm發(fā)光二極管(LED)激光器照射。采用細(xì)胞計(jì)數(shù)試劑盒(CCK-8)法檢測(cè)ALA-PDT對(duì)瘢痕成纖維細(xì)胞增殖的影響,并應(yīng)用實(shí)時(shí)聚合酶鏈反應(yīng)(real-time PCR)和Western blot檢測(cè)各組瘢痕成纖維細(xì)胞中膠原蛋白Ⅲ(CollagenⅢ)m RNA水平和蛋白表達(dá)水平。結(jié)果與空白對(duì)照組及PBS組比較,CCK-8結(jié)果表明,ALA-PDT能抑制瘢痕成纖維細(xì)胞生長(zhǎng),其抑制作用隨著ALA濃度增加逐漸增強(qiáng),當(dāng)ALA濃度為0.500 mmol/L時(shí)抑制作用最明顯。RT-PCR和Western blot結(jié)果表明,隨著ALA濃度增大,CollagenⅢm RNA水平和蛋白表達(dá)水平逐漸下降,當(dāng)ALA溶液濃度達(dá)到0.500 mmol/L時(shí)下降至最低值。結(jié)論 ALA-PDT能有效抑制瘢痕成纖維細(xì)胞增殖,抑制作用在ALA濃度為0.500 mmol/L時(shí)最強(qiáng)。ALA-PDT可能是通過(guò)抑制瘢痕成纖維細(xì)胞中CollagenⅢ的合成,進(jìn)而抑制瘢痕疙瘩的形成。
