人臍帶Wharton’s Jelly間充質(zhì)干細(xì)胞治療老年肌肉衰減綜合征的實(shí)驗(yàn)研究

【摘要】:目的:利用人臍帶Wharton’s Jelly間充質(zhì)干細(xì)胞(h UCMSCs)移植治療老年肌肉衰減綜合征模型小鼠,探討干細(xì)胞移植治療的方法及可行性。方法:利用后肢懸提技術(shù)建立老年肌肉衰減綜合征模型。取24月齡C57BL/6雄性小鼠24只,隨機(jī)分為靜脈注射干細(xì)胞組、肌肉注射干細(xì)胞組、靜脈注射漢克氏液組、肌肉注射漢克氏液組,每組6只。用胰酶消化法與差速貼壁法培養(yǎng)h UCMSCs,傳代3次后分別利用腓腸肌局部和尾靜脈注射的方法進(jìn)行h UCMSCs移植治療。通過(guò)比較最大等距跖屈肌力、腓腸肌的肌肉濕重、肌纖維的橫截面積、I型肌肉纖維的占比等指標(biāo)對(duì)h UCMSCs移植的療效進(jìn)行評(píng)價(jià)。結(jié)果:1.胰酶消化與差速貼壁法培養(yǎng)h UCMSCs,可獲得穩(wěn)定傳代及足量的細(xì)胞,能夠保證實(shí)驗(yàn)的需求。2.后肢懸提2周導(dǎo)致老年小鼠的體重下降,小鼠重新行走并接受h UCMSCs移植8周后,體重仍繼續(xù)下降。3.后肢懸提可以顯著降低老年小鼠最大等距跖屈肌力,h UCMSCs移植可以顯著改善老年小鼠的最大等距跖屈肌力,局部肌肉內(nèi)h UCMSCs移植效果要好于尾靜脈注射移植。4.h UCMSCs移植顯著增加腓腸肌的濕重、肌纖維的橫截面積及腓腸肌肌肉組織中I型肌肉纖維的占比,局部肌肉注射移植同樣優(yōu)于尾靜脈注射移植。結(jié)論:1.hUCMSCs易于體外培養(yǎng)擴(kuò)增,可以通過(guò)體外注射進(jìn)行細(xì)胞移植2.hUCMSCs移植可顯著增加腓腸肌的濕重,并顯著改善老年小鼠的下肢肌力,提示h UCMSCs移植在老年肌肉衰減綜合征模型動(dòng)物體內(nèi)能夠起到一定的治療作用。
[Abstract]:Objective: to study the feasibility of transplantation of Wharton's Jelly mesenchymal stem cells from human umbilical cord to treat senile muscle attenuation syndrome model mice.Methods: the model of senile muscle attenuation syndrome was established by hind limb suspension technique.Twenty-four C57BL/6 male mice aged 24 months were randomly divided into three groups: intravenous stem cell group, intramuscular stem cell injection group, Hank's fluid group and Hank's fluid group.HUCMSCs were cultured with trypsin digestion and differential adherent method. After three passages, the local and caudal vein injection of gastrocnemius was used for the transplantation of h UCMSCs respectively.The effect of h UCMSCs transplantation was evaluated by comparing the maximum isometric flexor muscle strength, muscle wet weight of gastrocnemius muscle, and the ratio of muscle fiber cross-sectional area to type I muscle fiber.The result is 1: 1.By trypsin digestion and differential adherence method, stable passage and sufficient number of cells could be obtained.The hind limb suspension for 2 weeks resulted in the weight loss of the aged mice, and the weight loss continued to decrease 8 weeks after the mice walked again and received h UCMSCs transplantation for 8 weeks.The hind limb suspension could significantly reduce the maximal isometric flexor muscle strength of the aged mice and the maximum isometric flexor muscle strength of the aged mice could be significantly improved by the transplantation of h UCMSCs.The effect of local intramuscular h UCMSCs transplantation was better than that of tail vein injection. 4. H UCMSCs transplantation significantly increased the wet weight of gastrocnemius muscle, the cross sectional area of muscle fiber and the proportion of type I muscle fiber in gastrocnemius muscle tissue.Local intramuscular injection was also superior to caudal vein transplantation.Conclusion\\\%\%\%\%\%\%\%\%\%\%\%\%\%\%\%\%\%\%\%\%\%\%\%\%\% UCMSCs can be easily cultured and amplified in vitro. 2.hUCMSCs transplantation can significantlyThese results suggest that h UCMSCs transplantation can play a therapeutic role in senile muscle attenuation syndrome model animals.
