HIF-1α介導(dǎo)BMSCs復(fù)合PLGA修復(fù)大鼠顱骨標(biāo)準(zhǔn)骨缺損的研究

【摘要】:目的構(gòu)建低氧誘導(dǎo)因子(HIF)-1α介導(dǎo)骨髓間充質(zhì)干細(xì)胞(BMSCs)復(fù)合聚乳酸-聚乙醇酸共聚物(PLGA)支架材料血管化組織工程骨,觀察其在大鼠顱骨標(biāo)準(zhǔn)骨缺損中的修復(fù)效果。方法將目的基因HIF-1α轉(zhuǎn)染至BMSCs后,與支架材料PLGA復(fù)合,修復(fù)SD大鼠顱骨雙側(cè)直徑5 mm的標(biāo)準(zhǔn)骨缺損(n=18),隨機(jī)分為3組:實(shí)驗(yàn)組植入HIF-1α-BMSCs/PLGA復(fù)合體(n=6),對(duì)照組植入BMSCs/PLGA復(fù)合體(n=6),材料組僅植入PLGA支架材料(n=6)。術(shù)后8周處死大鼠取材,分別行大體觀察、X線片檢查和HE染色觀察缺損區(qū)骨形成情況。結(jié)果大體觀察、X線片檢查和HE染色均顯示實(shí)驗(yàn)組缺損區(qū)新骨形成量明顯大于對(duì)照組及材料組,差異有統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論體內(nèi)研究表明HIF-1α能夠介導(dǎo)BMSCs促進(jìn)骨組織形成,PLGA是理想的支架材料,用其構(gòu)建的血管化工程骨能夠有效修復(fù)骨缺損。
[Abstract]:Objective to construct hypoxia inducible factor (HIF) -1 induced by bone marrow mesenchymal stem cells (BMSCs) compound poly lactic acid glycolic acid (PLGA) scaffolds for tissue engineering of vascularized bone in the skull, observe the standard bone defect in rats. Methods the repair effect of HIF-1 gene transfection to BMSCs alpha PLGA, composite and scaffold materials, standard bone defect repair SD rat skull bilateral 5 mm diameter (n=18), were randomly divided into 3 groups: the experimental group was implanted with HIF-1 alpha -BMSCs/PLGA complex (n=6), the control group was implanted with BMSCs/PLGA complex (n=6), materials were implanted into PLGA scaffolds (n=6) after 8. Weeks the rats were killed, respectively, for gross observation, X-ray examination and HE staining bone formation. The gross observation, X-ray examination and HE staining showed that the experimental group defect new bone formation was significantly greater than that of the control group and material group, the difference was statistically significant (P0.05). Conclusion in vivo studies indicate that HIF-1 alpha can mediate BMSCs to promote bone formation, PLGA is an ideal scaffold material, and its construction of vascular chemical process bone can effectively repair bone defects.
