【摘要】：目的探討白花丹素對骨肉瘤細(xì)胞MG-63(簡稱骨肉瘤細(xì)胞)增殖、凋亡的影響,并探討其可能的機制。方法將對數(shù)生長期的骨肉瘤細(xì)胞隨機分為四組,5μmol/L組、10μmol/L組和20μmol/L組分別加入含5、10和20μmol/L白花丹素的培養(yǎng)基,正常對照組加入含0.1%DMSO的完全培養(yǎng)基。培養(yǎng)24 h,采用CCK-8法檢測細(xì)胞增殖抑制率,Hoechst33342染色后計算細(xì)胞凋亡率,流式細(xì)胞術(shù)檢測活性氧(ROS)水平,Western blotting法檢測Bcl-2、Bax蛋白相對表達(dá)量。結(jié)果對照組、5μmol/L組、10μmol/L組、20μmol/L組細(xì)胞增殖抑制率和凋亡率均逐漸升高,組間兩兩比較P均0.05。對照組和5μmol/L組、10μmol/L組、20μmol/L組ROS水平、Bax蛋白相對表達(dá)量均逐漸升高,Bcl-2蛋白相對表達(dá)量逐漸降低,組間比較P均0.05。結(jié)論白花丹素能夠抑制骨肉瘤細(xì)胞增殖、促進(jìn)其凋亡,并呈濃度依賴性;升高細(xì)胞內(nèi)ROS水平、激活線粒體途徑可能是其作用機制。
[Abstract]:Objective to investigate the effect of Baihuadan on proliferation and apoptosis of osteosarcoma cell line MG-63 (osteosarcoma cell line). Methods Osteosarcoma cells in logarithmic phase were randomly divided into four groups: 5 渭 mol/L group, 10 渭 mol/L group and 20 渭 mol/L group. The normal control group was cultured on a complete medium containing 0.1% DMSO for 24 h. The inhibition rate of cell proliferation was detected by CCK-8 assay and the apoptosis rate was calculated by Hoechst33342 staining. The level of reactive oxygen species (Ros) was detected by flow cytometry and the relative expression of Bcl-2P Bax protein was detected by Western blotting. Results the proliferation inhibition rate and apoptosis rate of control group (5 渭 mol/L), 10 渭 mol/L group (10 渭 mol/L) and 20 渭 mol/L group increased gradually. The relative expression of ROS protein in 10 渭 mol/L group and 10 渭 mol/L group of 5 渭 mol/L group increased gradually and decreased gradually in 20 渭 mol/L group, while the relative expression of Bcl-2 protein decreased gradually in the control group and 5 渭 mol/L group (P 0.05). Conclusion Bax protein can inhibit the proliferation of osteosarcoma cells. Promoting its apoptosis in a concentration-dependent manner, increasing the level of ROS and activating the mitochondrial pathway may be the mechanism of its action.
