高壓氧對(duì)模型大鼠顱腦損傷CAT、SOD、GSH-Px和Nrf2影響的研究

【摘要】:目的:研究高壓氧對(duì)模型大鼠顱腦損傷過氧化氫酶(CAT)、超氧化物歧化酶(SOD)、谷胱甘肽過氧化物酶(GSH-Px)和核因子E2相關(guān)因子2(Nrf2)的影響。方法:健康SD大鼠30只,體重250~300 g,4~5月齡,隨機(jī)分為3組:假手術(shù)組、模型組和高壓氧組,每組10只,參照Feeney自由落體沖擊造模法建立顱腦損傷大鼠模型。假手術(shù)組只給予手術(shù),不造成顱腦損傷;模型組和高壓氧組給予顱腦損傷處理;高壓氧組用高壓氧治療,1次/d,10 d。用ELISA法檢測(cè)腦組織CAT、SOD、GSH-Px和Nrf2含量,采用實(shí)時(shí)熒光定量PCR檢測(cè)腦組織CAT、SOD、GSH-Px和Nrf2 mRNA相對(duì)表達(dá)量,用Western Blot測(cè)定腦組織Nrf2胞質(zhì)蛋白和Nrf2核蛋白。結(jié)果:假手術(shù)組、模型組、高壓氧組之間的CAT、SOD、GSH-Px、Nrf2 mRNA水平均有差異(P0.05)。模型組腦組織CAT、SOD、GSHPx、Nrf2 mRNA水平低于假手術(shù)組(P0.05),而高壓氧組則高于模型組(P0.05)。假手術(shù)組、模型組、高壓氧組之間的腦組織CAT、SOD、GSH-Px和Nrf2核蛋白水平存在差異(P0.05),模型組低于假手術(shù)組(P0.05),高壓氧組高于模型組(P0.05)。Western Blot檢測(cè)顯示:與假手術(shù)組比較,模型組和高壓氧組腦組織細(xì)胞質(zhì)(核)中Nrf2蛋白表達(dá)量有明顯升高(P0.05),高壓氧組升高更明顯(P0.05)。結(jié)論:高壓氧對(duì)模型大鼠顱腦損傷CAT、SOD、GSH-Px和Nrf2表達(dá)具有調(diào)節(jié)作用,通過這些調(diào)節(jié)改善機(jī)體氧化性損傷。
[Abstract]:Objective: to study the effects of hyperbaric oxygen on catalase catalase (CAT), superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) and nuclear factor E2 related factor (Nrf2) in model rats with craniocerebral injury. The rats were randomly divided into three groups: sham operation group, model group and hyperbaric oxygen group, each group with 10 rats. The model of craniocerebral injury was established by Feeney free falling body impact method. Model group and hyperbaric oxygen group were treated with craniocerebral injury, hyperbaric oxygen group was treated with hyperbaric oxygen for 10 days. The contents of GSH-Px and Nrf2 in brain tissue were measured by ELISA method, and the relative expressions of GSH-Px and Nrf2 mRNA in brain tissue were detected by real-time fluorescence quantitative PCR. Nrf2 cytoplasmic protein and Nrf2 nuclear protein in brain tissue were measured by Western Blot. Results: sham operation group, model group, model group, The level of GSH-PxNrf2 mRNA in brain tissue of model group was lower than that of sham operation group (P 0.05), but that of hyperbaric oxygen group was higher than that of model group. There were significant differences in the levels of GSH-Px and Nrf2 in brain tissue between hyperbaric oxygen group (HBO) group and sham operation group (P 0.05). The results of Western Blot analysis of hyperbaric oxygen group were as follows: compared with sham-operation group, hyperbaric oxygen group was higher than model group (P 0.05). In the model group and hyperbaric oxygen group, the expression of Nrf2 protein was significantly increased in the cytoplasm (nucleus) of brain tissue, especially in the hyperbaric oxygen group. Conclusion: hyperbaric oxygen can regulate the expression of GSH-Px and Nrf2 in CATSODX after craniocerebral injury in the model rats, and the expression of GSH-Px in the hyperbaric oxygen group is even higher than that in the hyperbaric oxygen group. These adjustments improve the oxidative damage of the body.
