小劑量雌激素肌肉注射聯(lián)合骨髓間充質(zhì)干細(xì)胞移植修復(fù)脊髓損傷

【摘要】:背景:多種手段聯(lián)合應(yīng)用可針對(duì)不同靶點(diǎn)進(jìn)行有效干預(yù),發(fā)揮疊加或協(xié)同作用,已成為當(dāng)前脊髓損傷治療的新思路。目的:探討骨髓間充質(zhì)干細(xì)胞移植聯(lián)合應(yīng)用小劑量雌激素對(duì)大鼠脊髓損傷后神經(jīng)功能恢復(fù)及炎癥反應(yīng)的影響。方法:將70只雄性SD大鼠隨機(jī)分5組,假手術(shù)組(n=10)僅暴露脊髓,不做損傷處理;模型組、雌激素組、干細(xì)胞組、聯(lián)合組(每組15只)采用改良的Allen’s法建立脊髓損傷模型。干細(xì)胞組、聯(lián)合組造模后立即在損傷部位移植骨髓間充質(zhì)干細(xì)胞;雌激素組、聯(lián)合組造模后第1,24小時(shí)肌肉注射17β-雌二醇。造模后第1,3,5,7天,進(jìn)行BBB行為學(xué)評(píng)價(jià);造模后6,12,24,72 h,應(yīng)用ELISA法測(cè)定脊髓勻漿組織白細(xì)胞介素1β、腫瘤壞死因子α水平,TUNEL染色觀察脊髓組織切片神經(jīng)細(xì)胞凋亡情況。結(jié)果與結(jié)論:(1)與假手術(shù)組比較,模型組造模后不同時(shí)間點(diǎn)的BBB行為學(xué)評(píng)分下降(P0.05);與模型組比較,干細(xì)胞、雌激素組、聯(lián)合組造模后5,7 d的BBB行為學(xué)評(píng)分升高(P0.05),且聯(lián)合組高于干細(xì)胞、雌激素組(P0.05);(2)與假手術(shù)組比較,模型組造模后6,12,24 h的白細(xì)胞介素1β、腫瘤壞死因子α水平升高(P0.05);與模型組比較,干細(xì)胞、雌激素組、聯(lián)合組造模后12,24 h的白細(xì)胞介素1β、腫瘤壞死因子α水平降低(P0.05),且聯(lián)合組低于干細(xì)胞、雌激素組(P0.05);(3)造模后72 h,模型組神經(jīng)細(xì)胞凋亡率高于假手術(shù)組,干細(xì)胞、雌激素組、聯(lián)合組低于模型組(P0.05),且以聯(lián)合組最低(P0.05);(4)結(jié)果表明在脊髓損傷中,雌激素與骨髓間充質(zhì)干細(xì)胞聯(lián)合應(yīng)用具有協(xié)同作用,能更有效抑制神經(jīng)細(xì)胞凋亡,促進(jìn)脊髓功能恢復(fù)。
[Abstract]:Background: the combined use of multiple means can effectively intervene against different targets and play a superimposed or synergistic role. It has become a new idea to treat spinal cord injury. Objective: to investigate the effect of bone marrow mesenchymal stem cell transplantation combined with low dose estrogen on the recovery of nerve function and inflammatory response after spinal cord injury in 70 rats. Male SD rats were randomly divided into 5 groups. Model group, estrogen group, stem cell group, combined group (15 rats in each group) were used to establish spinal cord injury model with modified Allen's method. Bone marrow mesenchymal stem cells (BMSCs) were transplanted immediately after modeling in the combined group, and 17 尾 -estradiol was injected intramuscularly in the estrogen group at 1h after modeling. The behavior of BBB was evaluated by BBB. The expression of IL-1 尾 and TNF- 偽 levels in spinal cord homogenate tissue were determined by ELISA method and Tunel staining was used to observe the apoptosis of neurons in spinal cord tissue slices. Results and conclusion: 1) compared with sham operation group, the expression of IL-1 尾 and TNF- 偽 in spinal cord homogenate tissue were determined by ELISA method. Compared with the model group, the BBB behavioral score of stem cell, estrogen group and combination group increased P0.05 at 7 days after modeling, and higher in the combined group than that in the stem cell group, and compared with the model group at different time points after the model making, the BBB behavioral score of the model group was decreased at different time points (P0.05), and that of the combined group was higher than that of the stem cell group. Compared with sham-operated group, the levels of IL-1 尾 and TNF- 偽 in model group increased at 1224 h after modeling, compared with model group, compared with model group, stem cells and estrogen group. The levels of IL-1 尾 and TNF- 偽 in the combined group were significantly lower than those in the control group (P 0.05) and the apoptotic rate of neurons in the model group was higher than that in the sham operation group, stem cell group and estrogen group at 72 h after the establishment of the model. The results showed that the combination of estrogen and bone marrow mesenchymal stem cells had synergistic effect on spinal cord injury, which could effectively inhibit neuronal apoptosis and promote the recovery of spinal cord function.
