脊柱結(jié)核不同病灶組織總RNA量的比較研究

【摘要】:研究目的:觀察脊柱結(jié)核不同病灶組織中總RNA的產(chǎn)量和質(zhì)量。實(shí)驗(yàn)研究方法:30例脊柱結(jié)核患者均行徹底清除病灶術(shù),術(shù)后用無菌的EP管收集患者的膿液、死骨、肉芽、病灶壁、硬化骨(所有組織標(biāo)本均用無菌剪刀、咬骨剪等器械剪碎),立即選取各種組織標(biāo)本50mg液氮冷凍后用研缽徹底研碎,加入Trizol試劑1ml進(jìn)行總RNA的提取;再選取30例冰凍一年的脊柱結(jié)核不同病灶組織標(biāo)本,也用同樣的方法進(jìn)行總RNA的提取,應(yīng)用ND-100分光光度儀檢測(cè)不同病灶組織中的總RNA的產(chǎn)量和質(zhì)量,同時(shí)用1%的瓊脂糖凝膠電泳分別檢測(cè)所提取總RNA的完整性。實(shí)驗(yàn)結(jié)果:新鮮的脊柱結(jié)核不同病灶組織標(biāo)本中,膿液組織中含有的總RNA量大且質(zhì)量好,其次為肉芽組織,含有總RNA量最少的為硬化骨,所有提取的總RNA經(jīng)瓊脂糖凝膠電泳以后條帶清晰完整且28S:18S的亮度為2:1。冰凍一年的不同病灶組織標(biāo)本中膿液組織的總RNA量最大且各種組織的總RNA經(jīng)瓊脂糖凝膠電泳后完整且28S為18S亮度的2倍,部分組織有少量的5S條帶。實(shí)驗(yàn)結(jié)論:新鮮的脊柱結(jié)核不同病灶組織標(biāo)本中膿液中含有的總RNA量大且質(zhì)量好,而且完整性良好,完全可以滿足后續(xù)的實(shí)驗(yàn)要求;冰凍一年的病灶組織標(biāo)本有少量的RNA發(fā)生降解;與新鮮組織標(biāo)本中總RNA量相比,冰凍一年的病灶組織標(biāo)本同樣可以用于后續(xù)的實(shí)驗(yàn)研究。
[Abstract]:Objective: to observe the yield and quality of total RNA in different lesions of spinal tuberculosis. The sepsis, dead bone, granulation, lesion wall and sclerosed bone were collected with sterile EP tube after operation (all tissue specimens were shredded with sterile scissors, bone biting scissors and other instruments). All kinds of tissue samples were frozen with 50mg liquid nitrogen and then ground thoroughly with mortar. The total RNA was extracted by adding 1ml of Trizol reagent. In addition, 30 specimens of different lesions of spinal tuberculosis were collected from 30 cases of frozen spinal tuberculosis for one year, and the total RNA was extracted by the same method. The output and quality of total RNA in different lesion tissues were detected by ND-100 spectrophotometer. At the same time, the integrity of total RNA was detected by 1% agarose gel electrophoresis. Results: the total RNA in the tissue of fresh spinal tuberculosis was high and the quality was good. The next was granulation tissue, and the sclerosing bone had the least total RNA. All the total RNA extracted by agarose gel electrophoresis were clear and complete, and the brightness of 28s: 18s was 2: 1. 1. The total RNA content of the pus tissue was the largest and the total RNA of various tissues was complete after agarose gel electrophoresis, and 28s was twice as bright as 18s. There were a few 5s bands in some tissues. Conclusion: the total RNA in the abscess of fresh spinal tuberculosis tissues is large, good quality and good integrity. It can completely meet the requirements of subsequent experiments. A small amount of RNA was degraded in frozen tissue samples for one year. Compared with the total amount of RNA in fresh tissue samples, frozen tissue samples for one year can also be used in subsequent experimental studies.
