Zn(Ⅱ)配位脂質(zhì)體用于高效且血清耐受的基因轉(zhuǎn)染(英文)

近年來,陽離子聚合物、陽離子脂質(zhì)體等非病毒載體在基因遞送中的應(yīng)用越來越受到人們的關(guān)注.然而,血清的存在會降低它們的轉(zhuǎn)染效率,從而阻礙它們在臨床中的使用.因此,非常有必要開發(fā)能抵抗血清干擾的非病毒基因載體.本工作合成了一種新型的鋅離子配位的脂質(zhì),其與1, 2-二油酰-SN-甘油-3-磷酰乙醇胺脂質(zhì)(DOPE)共自組裝形成陽離子脂質(zhì)體(Zn-DTAc脂質(zhì)體).實(shí)驗(yàn)結(jié)果表明,Zn-DTAc脂質(zhì)體可以有效地壓縮質(zhì)粒形成納米復(fù)合物.即使在50%胎牛血清存在的情況下,該復(fù)合物也能進(jìn)行有效的細(xì)胞內(nèi)化和內(nèi)涵體逃逸,并實(shí)現(xiàn)高效的基因轉(zhuǎn)染.
