胰島素在不同材料表面的聚集研究

蛋白的淀粉樣纖維化聚集與多種疾病密切相關(guān),如阿爾茲海默病等,因?yàn)榈鞍拙奂傻睦w維是潛在的納米材料,引起了大量科學(xué)家的興趣,并對(duì)此進(jìn)行研究。此前大部分研究都是在液態(tài)環(huán)境中進(jìn)行的。近年來(lái),研究者們意識(shí)到生物體內(nèi)的蛋白纖維化行為主要發(fā)生在細(xì)胞膜上,是一種在界面上發(fā)生的行為;另一方面,界面的性質(zhì)可以調(diào)控其纖維化聚集的形貌。本文中,我們主要做了以下幾個(gè)工作:(1)利用原子力顯微鏡觀察了胰島素蛋白在液態(tài)環(huán)境中的纖維化聚,發(fā)現(xiàn)胰島素蛋白先聚集成單體,進(jìn)一步形成初級(jí)纖維,初級(jí)纖維之間互相纏繞成成熟的纖維。我們還觀察了溫度的影響,發(fā)現(xiàn)溫度對(duì)其纖維化過(guò)程具有促進(jìn)作用。(2)利用原子力顯微鏡觀察了高濕度時(shí)胰島素在不同材料表面的聚集。在高濕度環(huán)境下,襯底表面會(huì)形成一層納米級(jí)的水膜,牛胰島素分子會(huì)在水膜中擴(kuò)散、遷移、聚集。在MICA表面形成排列比較規(guī)則的纖維;在APS-MICA表面上呈現(xiàn)出較雜亂的聚集狀態(tài);在高溫?zé)峤馐℉OPG)表面蛋白分子主要吸附在HOPG的臺(tái)階上,堆積成高點(diǎn)并沿著臺(tái)階呈線狀排列;在OTS-MICA表面,部分區(qū)域出現(xiàn)團(tuán)聚,小部分聚集成纖維狀。(3)為了進(jìn)一步研究影響胰島素在MICA表面聚...
