納米殼聚糖骨形成蛋白水凝膠修復(fù)下頜骨缺損實驗研究

【摘要】:目的:觀察可注射性納米殼聚糖骨形成蛋白水凝膠修復(fù)頜骨缺損的效果。方法:以納米殼聚糖為載體,加入重組人骨形成蛋白-2制備成可注射性納米殼聚糖骨形成蛋白水凝膠(NCS/BMP-2 Gel)。取9只SD大鼠,在雙側(cè)背部皮下分別注射200μl(100μg/ml)NCS/BMP-2 Gel及單純納米殼聚糖水凝膠(NCS Gel)。于術(shù)后10、20、30 d分別處死3只,取材進行大體及常規(guī)病理學(xué)觀察;選取54只SD大鼠,隨機均分為3組(n=18)制備下頜骨缺損模型,實驗組骨缺損中注入NCS/BMP-2Gel、對照組注入NCS Gel、空白組不注射。術(shù)后3、6、9周各組處死6只大鼠。分離下頜骨,用鉬靶掃描圖像,Image Pro-Plus軟件比較其頜骨缺損面積;常規(guī)病理觀察頜骨成骨情況。結(jié)果:實驗側(cè)、對照側(cè)均形成皮下結(jié)節(jié),實驗側(cè)30 d結(jié)節(jié)內(nèi)可見類骨樣結(jié)構(gòu);鉬靶掃描檢查:植入后3、6、9周時,實驗組剩余骨缺損面積均低于對照組及空白組(P0.05);組織學(xué)觀察顯示,實驗組下頜骨成骨情況優(yōu)于對照組及空白組。結(jié)論:NCS/BMP-2水凝膠有良好的生物相容性,能促進頜骨缺損修復(fù)。
