【摘要】：目的研究自制的納米銀豬小腸黏膜下層(nano-silver modified porcine small intestinal submucosa,NS-PSIS)生物腹壁修復材料的生物安全性。方法取新鮮豬小腸,采用機械處理(去除漿膜層、黏膜層及肌層)、化學SDS法脫細胞制備豬小腸黏膜下層生物材料(porcine small intestinal submucosa,PSIS),配制50μg/mL的單體納米銀(直徑15nm)溶液,將制備好的PSIS置入單體納米銀溶液中,制備成NS-PSIS,通過熱原試驗、皮膚刺激試驗、皮內刺激試驗、急性全身毒性試驗及體內銀吸收代謝試驗對自制的納米銀生物腹壁修復材料進行生物安全性評價。結果納米銀生物腹壁修復材料無熱原、皮膚刺激、皮內刺激及急性全身毒性�；鹧嬖游辗止夤舛确@示納米銀生物腹壁修復材料不造成血液及肝、腦、腎等重要器官的銀沉積。結論納米銀生物腹壁修復材料具有良好的生物安全性。
[Abstract]:Objective to study the biosafety of self-made nano-silver pig small intestinal submucosa (nano-silver modified porcine small intestinal submucosa,NS-PSIS) biological abdominal wall repair material. Methods the fresh pig small intestine was mechanically treated (removal of serosa, mucous membrane and muscle layer), the submucous biomaterial (porcine small intestinal submucosa,PSIS of pig small intestine was prepared by chemical SDS method, and the monomer silver nanoparticles (diameter 15nm) solution of 50 渭 g / mL was prepared. The prepared PSIS was put into the monomer silver nanoparticles solution, and the NS-PSIS, was prepared by pyrogen test, skin irritation test and intradermic stimulation test. Acute systemic toxicity test and in vivo silver absorption and metabolism test were used to evaluate the biosafety of self-made nano-silver biological abdominal wall repair materials. Results there were no pyrogen, skin irritation, intradermic irritation and acute systemic toxicity of nano-silver biological abdominal wall repair materials. Flame atomic absorption spectrophotometry showed that nano-silver biological abdominal wall repair materials did not cause silver deposition in blood, liver, brain, kidney and other important organs. Conclusion Nano-silver biological abdominal wall repair material has good biosafety.
