不同載銀量載銀羥基磷灰石細(xì)胞相容性與成骨活性的對(duì)比研究

【摘要】:目的·制備不同載銀量的載銀羥基磷灰石(Ag-HAp),探討不同載銀量Ag-HAp的細(xì)胞相容性及其對(duì)體外培養(yǎng)大鼠骨髓間充質(zhì)干細(xì)胞(BMSC)成骨活性的影響,確定適宜載銀量。方法·制備不同載銀量(x_(Ag)值)的Ag-HAp(x_(Ag)0.3、x_(Ag)0.5、x_(Ag)0.7)并對(duì)其進(jìn)行表征。將材料分為對(duì)照組(x_(Ag)0)和實(shí)驗(yàn)組(x_(Ag)0.3、x_(Ag)0.5、x_(Ag)0.7),檢測(cè)各組材料對(duì)BMSC的細(xì)胞相容性、成骨相關(guān)基因的表達(dá)和堿性磷酸酶分泌情況的影響。結(jié)果·材料表征結(jié)果證實(shí)合成產(chǎn)物為Ag-HAp,最終載銀量x_(Ag)0.7x_(Ag)0.5x_(Ag)0.3。細(xì)胞相容性實(shí)驗(yàn)結(jié)果顯示:x_(Ag)0.3-HAp和x_(Ag)0.5-HAp為合格,x_(Ag)0.7-HAp有輕微毒性。Real-time PCR結(jié)果顯示:與對(duì)照組比較,x_(Ag)0.3組和x_(Ag)0.5組成骨相關(guān)基因表達(dá)上調(diào),x_(Ag)0.7組成骨相關(guān)基因表達(dá)下調(diào)或無(wú)明顯改變。堿性磷酸酶定性定量實(shí)驗(yàn)顯示:x_(Ag)0.3組和x_(Ag)0.5組堿性磷酸酶活性均高于對(duì)照組,x_(Ag)0.7組堿性磷酸酶活性低于對(duì)照組。結(jié)論·改變?cè)牧现秀y的加入量(即改變x_(Ag)值)可制得不同載銀量Ag-HAp,當(dāng)x_(Ag)為0.3、0.5時(shí),其細(xì)胞毒性實(shí)驗(yàn)結(jié)果為合格,且可促進(jìn)大鼠BMSC成骨相關(guān)基因的表達(dá)及堿性磷酸酶的分泌。
[Abstract]:Objective to prepare silver-loaded hydroxyapatite (Ag-HAp) with different silver loading, to investigate the cellular compatibility of different silver-loaded Ag-HAp and its effect on (BMSC) osteogenic activity of rat bone marrow mesenchymal stem cells cultured in vitro, and to determine the suitable silver loading. Methods Ag-HAp (x _ (Ag) 0.3, x _ (Ag) 0.5, x _ (Ag) 0.7) with different silver loading (x _ (Ag) value) was prepared and characterized. The materials were divided into control group (x _ (Ag) 0) and experimental group (x _ (Ag) 0.3, x _ (Ag) 0.5, x _ (Ag) 0.7). The cellular compatibility of each group to BMSC was detected. Effect of osteogenic related gene expression and alkaline phosphatase secretion. Results the results of material characterization confirmed that the synthesized product was the final silver carrying capacity of Ag-HAp, 0.7x( Ag) 0.5x( Ag) 0.3. The results of cell compatibility test showed that x _ (Ag) 0.3-HAp and x _ (Ag) 0.5-HAp were qualified and x _ (Ag) 0.7-HAp was slightly toxic. Real-time PCR results showed that compared with the control group, The expression of bone-related genes in x( Ag) group was up-regulated, while the expression of bone-related genes in x( Ag) group was down-regulated or had no significant change. The results of qualitative and quantitative experiments showed that the alkaline phosphatase activity in x( Ag) group and x( Ag) group was higher than that in control group, and the alkaline phosphatase activity in x( Ag) group was lower than that in control group. Conclusion different silver loading Ag-HAp, can be prepared by changing the amount of silver in raw materials (that is, changing the value of x _ (Ag). When the x _ (Ag) is 0.3,0.5, the cytotoxicity test results are qualified. It can promote the expression of osteogenic related genes and the secretion of alkaline phosphatase in rats with BMSC.
