不同劑量氯化鑭對(duì)氧化鋁陶瓷顆粒誘導(dǎo)RAW264.7細(xì)胞產(chǎn)生炎癥反應(yīng)的保護(hù)作用
[Abstract]:Aim to observe the effect of different doses of lanthanum chloride (LaCl3) on the inflammatory response of mouse macrophage RAW264.7 induced by alumina (Al2O3) ceramic particles, and to provide a theoretical basis for studying the role of LaCl3 in aseptic loosening induced by Al2O3 ceramic particles. Methods Mouse macrophage RAW264.7. was cultured in vitro. According to the different culture medium, they were divided into 8 groups: blank control group (without LaCL3 and Al2O3 in the culture medium), Al2O3 group (containing 1 mg/mL Al2O3), LaCl3 Al2O3 group (2.5100 渭 mol / L LaCl3 after adding 1 mg/mL Al2O3) and LaCl3 group (containing 2.5100 渭 mol / L LaCl3). The expression of IL-1 尾, TNF- 偽, NF- 魏 B mRNA and protein were detected by ELISA,RT-PCR and Western blotting respectively. Results the results of ELISA showed that the protein secretion of IL-1 and TNF- 尾 in Al2O3 group and 100 渭 mol / L LaCl3 group were significantly higher than those in blank control group (P0.05). Compared with Al2O3 group, Al2O3 2.5 渭 mol / L LaCl3 group and Al2O3 10 渭 mol / L LaCl3 group decreased significantly (P 0.05). The levels of IL-1 尾 and TNF- 偽 in LaCl3 group were significantly lower than those in 100 渭 mol / L LaCl3 group (P0.05). The results of RT-PCR and Western blotting showed that IL-1 尾 and TNF- 偽 were significantly lower than that of 100 渭 mol / L Western blotting group (P0.05). The expression trend of NF- 魏 B mRNA and NF- 魏 B protein was consistent with that of ELISA. Conclusion 10 渭 mol / L LaCl3 can inhibit the secretion of IL-1 尾 and TNF- 偽 in macrophages to a certain extent, and the inflammatory inhibition of Lacl 3 may be related to the inhibition of NF- 魏 B signal transduction pathway.
