不同方法制備異種骨支架材料的生物相容性研究

【摘要】:目的:檢測3種不同方法制備異種骨支架材料的生物相容性。方法:通過急性毒性實(shí)驗(yàn)、熱源實(shí)驗(yàn)、皮內(nèi)刺激實(shí)驗(yàn)、皮下植入實(shí)驗(yàn)和細(xì)胞毒性實(shí)驗(yàn),初步評價(jià)經(jīng)過物理、化學(xué)及物理聯(lián)合化學(xué)不同處理方法制備的羊椎骨異種骨材料的生物相容性。結(jié)果:急性毒性實(shí)驗(yàn):物理、物理聯(lián)合化學(xué)和對照組動(dòng)物7d內(nèi)無毒性反應(yīng);化學(xué)組動(dòng)物初期出現(xiàn)呼吸困難輕度癥狀,,但無死亡情況。說明物理、物理聯(lián)合化學(xué)組材料無毒性;化學(xué)組材料有輕度毒性。熱源實(shí)驗(yàn):物理、化學(xué)及物理聯(lián)合化學(xué)組實(shí)驗(yàn)前后動(dòng)物體溫變化差值有統(tǒng)計(jì)學(xué)差異(P<0.05)。說明物理、物理聯(lián)合化學(xué)組材料不具有致熱源作用;化學(xué)組材料存在致熱源作用。皮內(nèi)刺激實(shí)驗(yàn):物理、物理聯(lián)合化學(xué)組注射處皮膚無明顯紅斑、水腫和壞死,說明物理、物理聯(lián)合化學(xué)組材料對皮膚無刺激性;化學(xué)組注射處皮膚在48、72h時(shí)出現(xiàn)極輕微紅斑,表明化學(xué)組材料有輕度刺激性。皮下植入實(shí)驗(yàn):組織學(xué)觀察發(fā)現(xiàn)物理聯(lián)合化學(xué)組材料周圍發(fā)現(xiàn)有少量成骨細(xì)胞規(guī)則排列,同時(shí)可見少量成骨,表明有異位成骨的趨勢;物理組和化學(xué)組材料周圍未見成骨細(xì)胞排列。細(xì)胞毒性實(shí)驗(yàn):MTT法證實(shí)物理、化學(xué)、物理聯(lián)合化學(xué)組材料浸液和對照組培養(yǎng)骨髓間充質(zhì)干細(xì)胞12d內(nèi),除物理、物理聯(lián)合化學(xué)組及對照組復(fù)合培養(yǎng)液的吸光度值比較無統(tǒng)計(jì)學(xué)差異(P>0.05)外,其他各組間均有統(tǒng)計(jì)學(xué)差異(P<0.05)。細(xì)胞能在物理、物理聯(lián)合化學(xué)組材料上良好地黏附、增殖、生長;細(xì)胞在化學(xué)組材料上生長受到抑制,材料孔隙內(nèi)無細(xì)胞黏附。結(jié)論:經(jīng)過物理聯(lián)合化學(xué)方法處理的異種骨材料具有良好的生物相容性;單純經(jīng)過物理或化學(xué)方法處理的異種骨材料生物相容性較差,不符合生物材料安全性標(biāo)準(zhǔn)。
[Abstract]:Objective: to investigate the biocompatibility of xenogeneic bone scaffolds prepared by three different methods. Methods: acute toxicity test, heat source test, intradermal stimulation test, subcutaneous implantation test and cytotoxicity test were carried out. Biocompatibility of different bone materials of sheep vertebrae prepared by chemical and physical methods. Results: acute toxicity test: physical, physical combined chemistry and control group had no toxic reaction within 7 days, the animals in the chemical group had mild symptoms of dyspnea at the initial stage, but there was no death. Physical and physical combination of chemical group materials are non-toxic; chemical group materials have mild toxicity. Heat source experiment: the difference of animal body temperature before and after physical, chemical and physical combined chemistry group had statistical difference (P < 0.05). The results show that the physical and physical chemical composition materials have no heat source effect, but the chemical composition materials have the heat source effect. Intradermal stimulation test: there was no obvious erythema, edema and necrosis in the injection site of physical and physical combined chemical group, which indicated that physical and physical combined chemical group had no irritation to the skin; The skin of the chemical group showed slight erythema at 48 h and 72 h, indicating that the chemical group had mild irritation. Subcutaneous implantation experiment: histological observation found that a small number of osteoblasts arranged regularly around the material in the physical combined chemical group, and a small amount of osteogenesis could be seen, indicating the tendency of heterotopic osteogenesis. No osteoblast arrangement was observed around the physical and chemical groups. Cytotoxicity test: MTT method confirmed physical, chemical, physical combined with chemical group material extract and control group cultured bone marrow mesenchymal stem cells within 12 days, except physics, There was no significant difference in the absorbance value between the physical combined chemical group and the control group (P > 0.05), but there was statistical difference among the other groups (P < 0.05). The cells could adhere, proliferate and grow well in the physical and physical groups, and the growth of the cells in the chemical group was inhibited, and there was no cell adhesion in the pores of the materials. Conclusion: the heterogenous bone materials treated by physical and chemical methods have good biocompatibility, and the heterogenous bone materials treated by physical or chemical methods alone have poor biocompatibility, which does not meet the safety standard of biomaterials.
