新型組織工程化改性膠原-殼聚糖復(fù)合支架制備與神經(jīng)細(xì)胞的相容性

【摘要】:背景:周?chē)窠?jīng)缺損的治療在再生醫(yī)學(xué)領(lǐng)域仍然具有挑戰(zhàn)性,支架材料的應(yīng)用被認(rèn)為是有前途的探索方向之一,理想的支架材料應(yīng)是安全、耐用和缺乏抗原性的。目的:探討制備一種新型的組織工程化改性膠原-殼聚糖支架,并探索其在大鼠橫斷坐骨神經(jīng)模型中用作橋梁修復(fù)神經(jīng)損傷的可能。方法:先制備出膠原-殼聚糖復(fù)合支架,然后用1-乙基-3-(3-二甲基氨基丙基)-碳化二亞胺,N-羥基琥珀酰亞胺對(duì)支架進(jìn)行化學(xué)改性即交聯(lián)。接著將神經(jīng)干細(xì)胞種植于支架上,之后連同支架-種子細(xì)胞在體內(nèi)植入坐骨神經(jīng)缺損部位。結(jié)果與結(jié)論:制得的變性交聯(lián)膠原-殼聚糖復(fù)合支架具有三維立體網(wǎng)狀結(jié)構(gòu)。將支架-種子細(xì)胞移植4周后,可在損傷的坐骨神經(jīng)中發(fā)現(xiàn)S-100神經(jīng)絲蛋白200表達(dá)。結(jié)果表明這種組織工程化改性交聯(lián)膠原-殼聚糖復(fù)合支架具有良好的細(xì)胞相容性。
[Abstract]:Background: the treatment of peripheral nerve defects is still challenging in regenerative medicine. The application of stent materials is considered to be one of the promising exploration directions. The ideal scaffold materials should be safe, durable and lack of antigenicity. Aim: to study the preparation of a new type of tissue engineering modified collagen chitosan scaffold and explore the possibility of using it as a bridge to repair nerve injury in rat sciatic nerve model. Methods: collagen-chitosan composite scaffolds were prepared, and then the scaffolds were chemically modified with 1-ethyl-3-(3-dimethylaminopropyl)-carbodiimide and N-hydroxysuccinimide. Neural stem cells were then implanted on the scaffold, and then together with the scaffold-seed cells, the sciatic nerve defect was implanted into the body. Results & conclusion: the modified cross-linked collagen-chitosan composite scaffolds have three-dimensional reticular structure. Four weeks after transplantation of scaffold-seeded cells, the expression of neurofilament 200 was found in injured sciatic nerve. 4 weeks after transplantation, the expression of neurofilament 200 was found in the injured sciatic nerve. The results showed that the cross-linked collagen-chitosan composite scaffolds with tissue engineering had good cellular compatibility.
