【摘要】：背景:目前骨缺損修復(fù)的主要方式是在缺損區(qū)充填骨替代材料,為了解骨缺損修復(fù)效果,需深入了解骨替代材料植入后骨缺損微細(xì)結(jié)構(gòu)改變。同步輻射顯微斷層成像技術(shù)(SR-μCT)可以在不用切片、染色的情況下,進(jìn)行高襯度分辨率、高空間分辨率的顯微三維成像,有極高的科研及臨床應(yīng)用價(jià)值。目的:探討在口腔醫(yī)學(xué)檢查中應(yīng)用SR-μCT對(duì)骨結(jié)合微米級(jí)檢查的可行性。方法:實(shí)驗(yàn)為隨機(jī)對(duì)照動(dòng)物實(shí)驗(yàn),在中國天津,天津市第五中心醫(yī)院完成。取24只雄性新西蘭大白兔制備下頜骨缺損模型,隨機(jī)分為4組,分別在缺損區(qū)植入自體骨、Bio-oss骨粉、β-磷酸三鈣骨粉,陰性對(duì)照組不植入任何材料。術(shù)后2,4,8周取材(包括缺損區(qū)及周圍部分正常骨組織),行SR-μCT檢查,之后制成組織切片行組織病理學(xué)檢查,從多角度觀察不同種類骨移植材料的骨修復(fù)情況。實(shí)驗(yàn)方案經(jīng)天津市第五中心醫(yī)院倫理委員會(huì)批準(zhǔn)。新西蘭大白兔的實(shí)驗(yàn)操作和取材遵循《關(guān)于善待實(shí)驗(yàn)動(dòng)物的指導(dǎo)性意見》規(guī)定,并與美國國立衛(wèi)生與健康研究院的指南一致。結(jié)果與結(jié)論:實(shí)驗(yàn)證實(shí)了SR-μCT能在不破壞樣品的情況下,完成對(duì)骨結(jié)合及新生骨微觀的觀察,得到骨體積、骨小梁數(shù)目及骨密度等的更準(zhǔn)確的定量數(shù)據(jù)。因此,通過SR-μCT能全面分析骨植入材料置入體內(nèi)后的生物相容性反應(yīng),深入了解不同種類骨移植材料在骨缺損處微米級(jí)結(jié)構(gòu)改變,可為提高骨缺損愈合的效果提供實(shí)驗(yàn)依據(jù)。
[Abstract]:Background: at present, the main method of bone defect repair is to fill the defect area with bone substitute material. In order to understand the effect of bone defect repair, it is necessary to deeply understand the microstructural changes of bone defect after bone replacement material implantation. Synchrotron radiation microtomography (SR- 渭 CT) is of great value in scientific research and clinical application because of its high contrast resolution and high spatial resolution. Objective: to investigate the feasibility of applying SR- 渭 CT to bone bonding micron examination in stomatology. Methods: the experiment was conducted in Tianjin, China, Tianjin Fifth Central Hospital. Twenty-four male New Zealand rabbits were randomly divided into 4 groups. Autogenous bone, Bio-oss bone powder, 尾 -tricalcium phosphate bone powder were implanted in the defect area, but no material was implanted in the negative control group. SR- 渭 CT was performed 8 weeks after operation, and then histopathological examination was performed to observe the bone repair of different kinds of bone graft materials from various angles. The experimental scheme was approved by the Ethics Committee of Tianjin Fifth Center Hospital. The experimental operation and sampling of New Zealand white rabbits follow the guidelines on the treatment of Experimental Animals and are consistent with the guidelines of the National Institutes of Health and Health of the United States. Results & conclusion: SR- 渭 CT was proved to be able to obtain more accurate quantitative data of bone volume, trabecular number and bone density without destroying the samples. Therefore, SR- 渭 CT can be used to analyze the biocompatibility of bone implant materials after implantation, and to understand the micron structure changes of different kinds of bone graft materials at the bone defect, which can provide experimental basis for improving the healing effect of bone defect.
