基于可降解磷酸酯鍵的生物材料和藥物緩釋體系的研究

【摘要】:磷酸酯鍵是一種廣泛存在于人體細胞和組織中的連接鍵,其不僅是卵磷脂的主要結(jié)構(gòu),更是核酸大分子DNA、RNA的組成部分。近年來,以磷酸酯鍵為連接鍵的材料的研究越來越受到科學(xué)工作者的重視,并證明其具有優(yōu)異的降解性和生物相容性,并廣泛的應(yīng)用于組織工程和藥物釋放領(lǐng)域。 鑒于磷酸酯鍵的諸多優(yōu)點,本論文設(shè)計并研究了以磷酸酯鍵為交聯(lián)鍵的熱固化可降解的生物彈性體材料。通過將低分子量聚乙二醇(PEG)進行環(huán)氧化和磷酸酯化修飾,按既定的比例將兩者混合均勻,在50℃下固化5小時,形成以磷酸酯為交聯(lián)鍵的三維網(wǎng)絡(luò)結(jié)構(gòu)交聯(lián)材料。實驗結(jié)果表明:(1)PEG的環(huán)氧化和磷酸化修飾制備簡單、產(chǎn)率高、轉(zhuǎn)化率高。(2)材料制備簡單且周期短。(3)材料的力學(xué)性能與預(yù)聚物結(jié)構(gòu)、配比等有直接關(guān)系。預(yù)聚物分子量越低,材料強度最高;且當P-OH基團和環(huán)氧基團比例為2:3時,材料強度最高。(4)材料具有良好的生物降解性,較低的細胞毒性和一定的藥物緩釋能力。 此外我們還設(shè)計了以磷酸酯鍵為連接鍵的藥物載體體系:以含有環(huán)氧基團的高分子材料或無機材料為載體,通過與含有P-OH官能團的藥物進行反應(yīng),制備以磷酸酯鍵為連接的載藥體系。通過紫外和高效液相色譜測試初步表明,構(gòu)建的兩種載藥體系都具有一定的藥物緩釋能力且具有一定藥物釋放選擇性。
[Abstract]:Phosphate bond is a kind of connective bond which widely exists in human cells and tissues. It is not only the main structure of lecithin but also the component of nucleic acid macromolecule DNA,RNA. In recent years, more and more attention has been paid to the study of phosphate bonding materials, which has been proved to have excellent biodegradability and biocompatibility, and has been widely used in tissue engineering and drug release fields. In view of the many advantages of phosphate bond, the thermo-degradable bioelastic material with phosphate bond as crosslinking bond was designed and studied in this paper.
