重力矢量及趨化誘導對脂肪干細胞在PLGA電紡膜上早期增殖的影響

【摘要】:觀察研究重力矢量方向及血小板衍生生長因子(PDGF)趨化誘導,在體外環(huán)境對人脂肪干細胞(ASCs)在PLGA電紡膜表面黏附和增殖活動的影響。體外原代培養(yǎng)從人頰脂墊提取的ASCs,根據(jù)不同重力矢量條件以及有無趨化因子PDGF將實驗分為4個組。A組:PDGF/重力矢量(+),含PDGF的PLGA電紡膜置于ASCs懸液底部;B組:PBS/重力矢量(+),空白(PBS)電紡膜置于ASCs懸液底部;C組:PDGF/重力矢量(-),含PDGF的PLGA電紡膜覆于ASCs懸液表面;D組:PBS/重力矢量(-),空白(PBS)電紡膜覆于ASCs懸液表面。分別于0、1、3、5、8、11d加入Alamar Blue繼續(xù)孵化24 h后測OD值,對照標準曲線,計算各組Alamar Blue的下降百分比,評估ASCs的黏附增殖活動。結果顯示:在重力矢量(+)組的早期(第1~4 d),無論趨化因子PDGF存在與否,Alamar Blue的下降百分比都能達到30%左右,不影響細胞的定植、黏附與增殖。但是在重力矢量(-)組,缺少趨化因子PDGF的Alamar Blue的下降百分比為25%左右,細胞將無法正常的定植與黏附。PDGF具有明顯誘導ASCs趨化聚集作用,能減輕不利重力因素的影響,顯著改善ASCs黏附與增殖。
[Abstract]:The effects of gravity vector direction and platelet-derived growth factor (PDGF) chemotaxis on the adhesion and proliferation of human adipose stem cell (ASCs) on the surface of PLGA electrospun membrane were studied in vitro. ASCs, extracted from human buccal fat pad in vitro was divided into four groups according to different gravity vector conditions and chemokine PDGF. Group A: PDGF/ gravity vector (), PLGA electrospun film containing PDGF at the bottom of ASCs suspension; group B: PBS/ gravity Vector (), blank (PBS) electrospun film is placed at the bottom of ASCs suspension, C group is PDGF/ gravity vector (-), PLGA electrospun film containing PDGF is coated on ASCs suspension surface, group D is PBS/ gravity vector (-), blank (PBS) electrospun film is coated on ASCs suspension surface. The values of OD were measured after incubating for 24 h after adding Alamar Blue for 24 hours. The percentage of Alamar Blue decreased in each group was calculated and the adhesion and proliferation of ASCs were evaluated. The results showed that in the early stage of gravity vector group (1 ~ 4 d), the percentage of, Alamar Blue decreased by about 30%, regardless of whether the chemokine PDGF existed or not, which did not affect cell colonization, adhesion and proliferation. However, in the gravity vector (-) group, the percentage of Alamar Blue in the absence of chemokine PDGF is about 25%, and the cells will not be able to colonize and adhere normally. PDGF can obviously induce the chemotactic aggregation of ASCs, which can reduce the influence of unfavorable gravity factors. The adhesion and proliferation of ASCs were significantly improved.
