基于咪唑—鋅離子作用的形狀記憶水凝膠的構(gòu)建及其作為管狀細(xì)胞支架的應(yīng)用探索

【摘要】:本文利用一步法以聚乙二醇二丙烯酸酯(PEGDA)為交聯(lián)劑,1-乙烯基咪唑和丙烯腈為單體共聚合成一種偶極-偶極增強(qiáng)水凝膠。該水凝膠的力學(xué)強(qiáng)度在微量鋅離子溶液中顯著提高,進(jìn)一步研究發(fā)現(xiàn)凝膠和少量金屬離子的絡(luò)合作用能夠固定住臨時(shí)形狀,去除離子可使凝膠恢復(fù)原始形狀,從而實(shí)現(xiàn)形狀記憶功能,而且這種記憶特性是可重復(fù)的。此外,此水凝膠有良好的細(xì)胞相容性,細(xì)胞能在該水凝膠上很好的生長(zhǎng)。將長(zhǎng)有細(xì)胞的水凝膠平片卷成管狀,然后放置在5mmol/L鋅離子培養(yǎng)基中固定形狀,放入細(xì)胞培養(yǎng)基中培養(yǎng)一段打開管狀凝膠支架發(fā)現(xiàn),細(xì)胞仍能很好的粘附在凝膠上并保持正常形態(tài)。這種包含在凝膠中的咪唑環(huán)配體和微量鋅離子的絡(luò)合-解絡(luò)合策略提供了一種實(shí)現(xiàn)形狀記憶凝膠的簡(jiǎn)單且普遍的方法,并且由于其實(shí)現(xiàn)過(guò)程對(duì)細(xì)胞沒(méi)有大的傷害,,很有希望作為一種可植入組織工程支架應(yīng)用于生物領(lǐng)域。
[Abstract]:A dipole-dipole reinforced hydrogel was prepared by one-step copolymerization of poly (ethylene glycol diacrylate) (PEGDA) as crosslinking agent and acrylonitrile as monomer. The mechanical strength of the hydrogel was significantly increased in trace zinc ion solution. It was found that the complexation of the gel with a small amount of metal ions could fix the temporary shape, and the removal of ions could restore the original shape of the gel. Thus the shape memory function is realized, and the memory property is repeatable. In addition, the hydrogel has good cell compatibility and cells can grow well on the hydrogel. The hydrogel flattens with long cells were rolled into tubules, then placed in 5mmol/L zinc ion medium to fix the shape, and cultured in the cell culture medium for a section of open tubular gel scaffold. Cells still adhere well to the gel and maintain normal morphology. This complexation-desorption strategy of imidazole ring ligands and trace zinc ions in the gel provides a simple and universal method for realizing shape memory gel, and because of its implementation process, it does not cause much damage to cells. It is promising to be used as an implantable scaffold for tissue engineering in biological field.
