膠原海綿復(fù)合人脂肪干細(xì)胞促血管新生

【摘要】:背景:研究發(fā)現(xiàn),脂肪干細(xì)胞膠原復(fù)合物可促進(jìn)脂肪干細(xì)胞向成熟脂肪細(xì)胞分化、成熟,并可促進(jìn)血管新生。目的:探查膠原海綿復(fù)合脂肪干細(xì)胞的生物學(xué)特性及其對血管新生的影響。方法:(1)實(shí)驗(yàn)組將人脂肪干細(xì)胞接種于膠原海綿上,以細(xì)胞單獨(dú)培養(yǎng)為陰性對照組,培養(yǎng)24 h后,檢測細(xì)胞黏附率;培養(yǎng)第2,4,6天,檢測細(xì)胞增殖與培養(yǎng)液中血管內(nèi)皮生長因子水平;(2)雞胚絨毛尿囊膜實(shí)驗(yàn):暴露孵育7 d的雞胚尿囊膜,分4組,空白組向尿囊膜加入0.2 mL無菌PBS,脂肪干細(xì)胞組加入0.2 mL 2×10~8 L~(-1)第3代脂肪干細(xì)胞懸液,膠原海綿組加入膠原海綿,復(fù)合組加入2×10~8 L~(-1)第3代脂肪干細(xì)胞懸液0.2 mL與膠原海綿。孵育7 d后,測定尿囊膜周圍微血管計(jì)數(shù)。結(jié)果與結(jié)論:(1)細(xì)胞黏附率:脂肪干細(xì)胞在膠原海綿上的黏附率為(93.04±0.67)%;(2)細(xì)胞增殖與培養(yǎng)液中血管內(nèi)皮細(xì)胞生長因子水平:實(shí)驗(yàn)組培養(yǎng)第6天的細(xì)胞增殖A值高于對照組(P0.01),培養(yǎng)第4,6天的血管內(nèi)皮生長因子水平高于對照組(P0.05);(3)雞胚絨毛尿囊膜實(shí)驗(yàn):脂肪干細(xì)胞組、膠原海綿組、復(fù)合組微血管計(jì)數(shù)高于空白組(P0.05),復(fù)合組高于脂肪干細(xì)胞組、膠原海綿組(P0.05);(4)結(jié)果表明:脂肪干細(xì)胞與膠原海綿具有良好的生物相容性,兩者復(fù)合有良好的促進(jìn)血管新生作用,其機(jī)制可能與促進(jìn)脂肪干細(xì)胞增殖與分泌血管內(nèi)皮生長因子相關(guān)。
[Abstract]:Background: it has been found that the collagen complex of adipose stem cells can promote adipose stem cells to differentiate into mature adipocytes and promote angiogenesis. Objective: to investigate the biological characteristics of collagen sponge combined with adipose stem cells and its effect on angiogenesis. Methods: (1) Human adipose stem cells were inoculated on collagen sponge in experimental group and cultured alone as negative control group. (2) Chorioallantoic membrane test: chorioallantoic membrane was exposed for 7 days and divided into 4 groups. The blank group added 0.2 mL aseptic PBSto the allantoic membrane, the adipose stem cell group added 0.2 mL 2 脳 10 8 L ~ (-1) third generation adipose stem cell suspension, the collagen sponge group added the collagen sponge, and the compound group added 2 脳 10 8 L ~ (-1) 2 脳 10 8 L ~ (-1) third generation adipose stem cell suspension 0.2 mL and collagen sponge. After incubation for 7 days, microvessel count around allantoic membrane was measured. Results and conclusion: (1) Adhesion rate of adipose stem cells on collagen sponge was (93.04 鹵0.67); (2) cell proliferation and vascular endothelial growth factor level in culture medium: the A value of cell proliferation in experimental group was higher than that in control group on the 6th day of culture. The level of vascular endothelial growth factor (VEGF) on the 4th day of culture was higher than that in the control group (P0.05); (3). In the collagen sponge group, the microvessel count in the composite group was higher than that in the blank group (P0.05), and that in the composite group was higher than that in the adipose stem cell group (P0.05). The results of collagen sponge group (P0.05); (4) showed that adipose stem cells had good biocompatibility with collagen sponge. The mechanism may be related to the proliferation of adipose stem cells and the secretion of vascular endothelial growth factor.
